Abstract
Angiotensin I Konversi Enzyme (ACE) Peptide diakui secara luas sebagai penyembuhan untuk hipertensi. Peptida bioaktif dari sumber alami, termasuk ikan laut, lebih dipertimbangkan karena tidak memiliki efek kerusakan. Tujuan dari penelitian ini adalah penyaringan keberadaan peptida penghambatan ACE potensial dari Protoamine Salmon. Peptida penghambatan ACE diperoleh dari protamin salmon setelah 16 jam hidrolisis oleh berbagai enzim dan diikuti oleh ultrafiltrasi pada 3 KDA memotong berat molekul (MWCO) membran. Urutan IR dianalisis dengan kromatografi cair tandem spektrometrik massa (LC-MS / MS). Aktivitas penghambatan ACE diukur menggunakan kromatografi cair performa tinggi fase terbalik (RP-HPLC). Sebagian besar peptida penghambatan ACE potensial adalah SYN berukuran menggunakan metode fase padat Peptide SYN SIS. Hasil menunjukkan bahwa trypsin hydrolyzate memiliki aktivitas penghambatan ACE tertinggi dibandingkan dengan o r hydrolysates dengan nilai IC50 135,96 μg / ml. LC-MS / MS analisis protamin tryptic mengidentifikasi dua puncak utama dengan tiga urutan peptida IV, ser-ser-arg-pro-arg (SR-6), ser-ser-ser-arg-pro-ile-arg ( SR-7), pro-arg-arg-ala-ser-arg (PR-6) yang bersumber dari Salmine of Chum Salmon (oncorhynchus keta). Bulu Rendah T kedua puncak difraksinasi dan uji aktivitas penghambatan ACE menunjukkan bahwa fraksi 2, milik Peptida SR-6 dan SR-7, memiliki aktivitas tertinggi. Nilai IC50 Peptide SR-7 adalah 94,98 ± 2,63 μm. Oleh karena itu, dapat disimpulkan bahwa protamine salmon memiliki peptida penghambatan ACE dan mungkin bermanfaat untuk mencegah hipertensi dan dapat digunakan sebagai bahan makanan fungsional.
English Abstract
Angiotensin I converting enzyme (ACE) inhibitory peptide is widely recognized as possible cures for hypertension. Bioactive peptides from natural sources, including marine fish, are more considered because it has no harm effects. objective of this study is screening presence of potential ACE inhibitory peptide from salmon protamine. ACE inhibitory peptide was obtained from salmon protamine after 16 hours of hydrolysis by various enzymes and followed by ultrafiltration on a 3 kDa molecular weight cut off (MWCO) membrane. ir sequences were analysed by Liquid Chromatography Tandem Mass Spectrometric (LC-MS/MS). ACE inhibitory activity was measured using Reverse Phase High Performa Liquid Chromatography (RP-HPLC). most potential ACE inhibitory peptide was syn sized using a method of Solid Phase Peptide Syn sis. results indicated that trypsin hydrolysate had highest ACE inhibitory activity compared to o r hydrolysates with IC50 value 135.96 μg/ml. LC-MS/MS analysis of tryptic protamine identified two major peaks with three IV peptide sequences, Ser-Ser-Arg-Pro-Ile-Arg (SR-6), Ser-Ser-Ser-Arg-Pro-Ile-Arg (SR-7), Pro-Arg-Arg-Ala-Ser-Arg (PR-6) which sourced from salmine of Chum Salmon (Oncorhynchus keta). Fur rmore se two peaks were fractionated and ACE inhibitory activity assay showed that fraction 2, belongs to SR-6 and SR-7 peptides, has highest activity. IC50 value of peptide SR-7 was 94.98 ± 2.63 μM. refore, it can be concluded that salmon protamine has ACE inhibitory peptides and it may be beneficial for preventing hypertension and can be used as functional food ingredients.
