Abstract

Pendahuluan: Diet minyak jelantah (DMJ) mengandung asam lemak trans yang terbentuk dari reaksi hidrogenasi parsial, oksidasi, dan pemanasan minyak kelapa sawit selama proses penggorengan. Komsumsi asam lemak trans jangka panjang meningkatkan stress oksidasi dan menyebabkan apoptosis sel β pankreas. Tujuan: Untuk mengetahui efek DMJ terhadap ekspresi F 2 -Isoprostanes dan apoptosis sel β pankreas tikus wistar in vivo. Metode: Penelitian eksperimental selama 8 minggu terhadap 18 tikus wistar yang terbagi secara acak dalam 3 grup (n=6 untuk tiap grup) yaitu kelompok diet normal (DN), diet a rogenik (DA), dan DMJ. Ekspresi F 2 -Isoprostanes diukur secara semikuantitatif dengan pewarnaan histokimia. Apoptosis sel- β dievaluasi dinilai dari pengkerutannya ( shrinkage ). Data dinyatakan dalam median (min-maks) untuk F 2 -isoprostanes; dan mean±SD untuk apoptosis sel- β. Analisa statistik menggunakan one-way ANOVA diikuti uji Tuckey untuk apoptosis sel- β dan Kruskal-Wallis diikuti uji Mann-Whitney untuk ekspresi F 2 -Isoprostanes. Hasil: Ekspresi F 2 -Isoprostanes meningkat pada DMJ dan DA dibandingkan dengan DN [33(28-39) dan 36(26-42) vs 3(3-9), p=0,002]. Apoptosis sel- β meningkat pada DMJ dan DA dibandingkan dengan DN [13,00±3,35 and 12,83±2,79 vs 2,33±1,21, p=0,000]. Tidak ada perbedaan bermakna pada DMJ dan DA terhadap ekspresi F 2 -Isoprostanes ataupun apoptosis sel- β [33,00(28 -39) vs 36(26-42), p=0,485; 13,00±3,35 vs 12,83±2,79, p=0,949]. Simpulan: Ekspresi F 2 -Isoprostanes dan apoptosis sel- β pank reas meningkat bermakna pada DMJ. Perlu penelitian lebih lanjut untuk menilai korelasi peningkatan ekspresi F 2 -isoprostanes dan apoptosis sel- β.

English Abstract

Background: Reused cooking oil diet (RCOD) has high trans fatty acid content as result of partial hydrogenation, oxidation, and rmal alteration of palm oil during frying. Chronic trans fat consumption will promote oxidative stress mechanism and lead to β -cell apoptosis. Objective: To examine effects of RCOD on F 2 -Isoprostanes expression and apoptosis of wistar rats pancreatic β -cell in vivo. Methods: Experimental study on wistar rats. 18 Wistar rats were devided into three groups (n=6 for each) randomly, fed with Normal Diet (ND), A rogenic Diet (AD), and RCOD respectively for 8 weeks. F 2 -Isoprostanes expression was examined semiquantitatively by immunohystochemistry. β -cell apoptosis was evaluated through its shrinkage, by Hematoxilen-Eosin stainning. Data were expressed in median and range for F 2 - isoprostanes; and mean±SD for β -cell apoptosis. statistical significance was assessed by one-way ANOVA followed by Tuckey post hoc test for β -cell apoptosis and Kruskal-Wallis followed by Mann-Whitney test for F 2 -Isoprostanes expression. Results: F 2 -Isoprostanes expression significantly increased in RCOD and AD compared to ND [33(28-39) and 36(26-42) vs 3(3- 9), p=0.002]. β -cell apoptosis significantly increased in RCOD and AD compared to ND [13.00±3.35 and 12.83±2.79 vs 2.33±1.21, p=0.000]. re were no significan differences between RCOD and AD on F 2 - Isoprostanes expression nor apoptosis of β -cell [33.00(28-39) vs 36(26-42), p=0.485; 13.00±3.35 vs 12.83±2.79, p=0.949]. Conclusion: F 2 -Isoprosta nes expression and pancreatic β -cell apoptosis significantly increased in RCOD compared to ND. Fur r investigation is needed to examine correlation between alteration of F 2 - isoprostanes and β -cell apoptosis in RCOD and AD.
