Abstract
Sambung (Blumea balsamifera) dan jahe (Zingiber officinale roscoe) terkenal sebagai tanaman obat di Asia dan Afrika. Tanaman tersebut juga secara tradisional digunakan untuk menurunkan motilitas usus dan menjadi agen antihipertensi. Penelitian ini dilakukan untuk mengevaluasi mekanisme kegiatan ekstrak metanol dari rimpang jahe dan daun sambung pada motilitas usus. Setelah ekstrak metanol herbal dipersiapkan, penelitian farmakologi pada usus halus marmut yang terisolasi yang dicatat dengan Powerlab Recorder. Kurva respon-dosis agonis dari asetilkolina, histamina, dan serotonin yang dosisnya mulai dari 1x10-6 sampai 3x10-8M dengan atau tanpa ekstrak metanol jahe diplot dan demikian pula dengan kurva respon-dosis dari Sambung yang dicampur dengan atau tanpa penyekat atropin atau promethazine (1x10-8M). Ekstrak metanol jahe (0.01, 0.1,1,10 and 100 ug/ml) bekerja sebagai aktifitas anti-muskarinik, anti-histaminik, dan anti-serotonin serta memberikan efek relaksasi usus halus. Ekstrak jahe tampaknya lebih bekerja sebagai antimuskarinik. Namun, lima dosis ekstrak metanol Sambung (125, 250, 500, 750 and 1000 μg/ml) meningkatkan pengerutan usus halus marmut. Hasil penelitian menunjukkan bahwa semua konsentrasi metanol jahe memiliki aktifitas antikolinergik (M3), anti-histamin (H1), dan anti-serotonin (5HT) dan condong secara kompetitif lebih menghalangi reseptor muskarinik; sebaliknya, Sambung lebih condong untuk mengaktifkan respetor muskarinik yang akhirnya mengarah ke kontraksi usus halus marmut.
English Abstract
Sambung (Blumea balsamifera) and ginger (Zingiber officinale roscoe) are well known as medicinal plants in Asia and Africa. y have also been traditionally used to decrease motility of intestine and be antihypertensive agents. present study is conducted to evaluate activity mechanism of methanolic extract of ginger rhizome and sambung leaves on intestine motility. After methanolic extracts of herbs were prepared, pharmacological study on isolated guinea pig ileum was recorded by Powerlab Recorder. Agonists dose-response curves of acetylcholine, histamine and serotonin whose doses were started from 1x10-6 to 3x10-8M with or without methanolic extract of ginger were plotted as well as dose-response curve of sambung with and without atropine or promethazine blockers (1x10-8M). methanolic extracts of ginger (0.01, 0.1,1,10 and 100 ug/ml) worked as anti-muscarinic, anti-histaminic and antiserotogenic activities and y gave ileum relaxation effect. ginger extract is more likely to work as antimuscarinic. However, five doses of sambung methanolic extract (125, 250, 500, 750 and 1000 μg/ml) increased contractility of guinea pig`s ileum. results indicate all methanolic concentrations of ginger have antimuscarinic (M3), antihistaminic (H1) and anti-serotonergic (5HT) activities and y are more likely to inhibit muscarinic receptors competitively; whereas, sambung is more likely to activate muscarinic receptors eventually leading to guinea pig`s ileum contraction.
