Abstract

MMP-9 merupakan enzim khusus yang berperan dalam degradasi extracellular matrix (ECM) pada proses angiogenesis. MMP-9 meregulasi neovaskularisasi melalui mekanisme remodeling kapiler membran basement. Diantara isoflavones, genistein diketahui merupakan inhibitor pertumbuhan sel kanker yang poten, serta mempengaruhi proliferasi sel endotel vaskular dalam neovaskularisasi kornea. Penelitian ini bertujuan untuk mengetahui efek genistein dalam ekstrak metanol kacang tunggak (Vigna unguiculata) terhadap ekspresi MMP-9 kornea model inflamasi pada tikus strain Wistar. Desain penelitian ini adalah interventional experimental study. Enam puluh tiga ekor tikus dipilih secara acak, 3 tikus sebagai kontrol, 60 tikus diinduksi dengan NaOH 1 M (sebagai model inflamasi) dan diberikan genistein dari ekstrak metanol kacang tunggak dengan dosis 25 ;M, 50 ;M dan 100 ;M sebanyak empat kali sehari. Kadar genistein diukur dengan metode Kromatografi Cair Kinerja Tinggi (KCKT). Induksi inflamasi menggunakan kertas saring yang dibasahi NaOH 1 M dan diletakkan pada puncak kornea mata kanan selama 60 detik digunakan sebagai kontrol positif dan ekstrak metanol kacang tunggak diberikan setelah induksi NaOH. Ekspresi MMP-9 diobservasi pada jam ke 6, 24, 48, 96, dan 168 menggunakan metode imunohistokimia. Masing-masing dosis ekstrak metanol kacang tunggak menurunkan ekspresi MMP-9 secara signifikan (p=0.000). Interaksi antara waktu pemberian dan dosis mempengaruhi ekspresi MMP-9 (R square (r2) = 36.7%). Regresi linear antara ekspresi MMP-9 dan dosis ekstrak metanol kacang tunggak menghasilkan estimated effective dose (ED) sebesar 89.9456 ;M dengan persamaan Y = 20.827-0.098X. Genistein dalam ekstrak metanol kacang tunggak menurunkan ekspresi MMP-9 pada kornea model inflamasi tetapi lama pemberian tidak berpengaruh.

English Abstract

Matrix metalloproteinase-9 (MMP-9), a specialized enzymes capable of proteolytically degrading extracellular matrix proteins, have been postulated to play an important role in angiogenesis. It has been suggested that MMP-9 can regulate neovascularization using mechanisms o r than simple remodeling of capillary basement membrane. Among isoflavones, genistein has been reported to be most potent inhibitor of cancer cell growth in vitro but also impair proliferation of vascular endo lial cells in corneal neovascularization. In this study,we evaluate effect of genistein in cowpea (Vigna unguiculata) methalonic extract on MMP-9 expression in rat corneal-alkali induced inflammation. An interventional experimental study, sixty three rats were randomly choosen, 3 rats as control, 60 rats were induced with NaOH 1M (as inflammation models) and administered with 25 ;M, 50 ;M and 100 ;M of genistein from cowpea methanolic extract four times daily. Genestein composition measured by High Performance Liquid Chromatography (HPLC) methode. Alkali burn as inflammation model using NaOH 1M infiltration through filter paper which were aplicated on center cornea of right eye for 60 seconds as positive control and cowpea methalonic extract were administered immediately after alkali burn by NaOH. MMP-9 expression was observed at 6, 24, 48, 96, and 168 hours by immunohistochemistry. Each dose of cowpea methalonic extract significantly decreased MMP-9 expression (p=0.000). Interaction between time of administration and dose of administration had influenced MMP-9 expression (R square (r2) = 36.7%). linear regression between MMP-9 expression and administration dose resulted in estimated effective dose (ED) 89.9456 ;M with Y = 20.827-0.098X. Genistein in cowpea methalonic extract decreased MMP-9 expression on NaOH alkali burn corneal inflammation in rats but time of administration not affect MMP-9 expression.
