Abstract

Epidermal growth factor (EGF) is a growth factor that is thought to have a function in the reproduction system. One of them is accelerating the maturity of oocyte. The purpose of this research is to investigate characterization of EGF in carp (Cyprinus carpio) and determine the expression of EGF in various tissues and the early stages of development on fish (eggs to larvae). Epidermal growth factor (EGF) cDNA was cloned from ovary by a reverse transcription polymerase chain reaction and confirm to get sequence. The similarity of homologous sequences of 81.5% against the zebra fish. However, unfortunately on this research still not found full length on carp EGF, so at next treatment of the test expression using only partial sequences. Different tissues on carp, were tested using real time PCR to quantify the expression of EGF. The results showed, almost all of the tissues to be tested did not have a significant difference. Other treatment conducted by observing the EGF expression in ovarian development stage which had iii been injected with HCG, the using of egg and larval stages of development. Similar results are shown on all treatments, there is no significant difference on any observations. One possible reason might be EGF has a long sequence and from the EGF family. Whereas, in this study, only get partial sequences, so that the primer might be not specific and the expression shown in the data may be derived from more than one EGF on carp.
