Abstract
Inflammatory Bowel Disease (IBD) adalah penyakit inflamasi kronik pada saluran pencernaan terutama usus halus dan usus besar. Salah satu faktor penyebab IBD adalah gangguan respon imun yang dapat dirangsang oleh adanya agen bakteri, virus dan xenobiotik. Salah satu penyebab gangguan respon imun dalam usus halus adalah indometasin yang termasuk dalam NSAID ( non-steroidal anti-inflammatory drug ). Indometasin dapat menstimulasi produksi ROS sehingga diperlukan antioksidan pada terapi IBD. Penelitian ini bertujuan untuk menjelaskan potensi ekstrak polifenol rumput laut coklat ( Sargassum duplicatum Bory) dalam menurunkan kadar malondialdehid (MDA) dan aktivitas protease, memperbaiki ekspresi protein ZO-1 dan occludin serta memperbaiki kerusakan jaringan jejunum usus halus tikus yang dipapar indometasin. Penelitian ini dibagi menjadi dua tahap. Tahap pertama adalah ekstraksi senyawa polifenol rumput laut coklat, uji fitokimia, kromatografi lapis tipis (KLT) preparatif dan analisis senyawa ekstrak dengan spektrofotometri inframerah (IR). Tahap kedua adalah pengujian ekstrak polifenol rumput laut coklat ( Sargassum duplicatum Bory) terhadap tikus IBD. Penelitian ini menggunakan hewan coba tikus putih ( Rattus norvegicus ) jantan galur wistar. Kelompok tikus sakit (IBD) diinduksi indometasin dengan dosis 15 mg/kg BB satu kali secara oral dan diinkubasi selama 7 hari. Tikus dinyatakan IBD setelah 24 jam pemaparan indometasin yang diketahui dari kerusakan mukosa (vili) jejunum usus halus berdasarkan pengamatan gambaran histologisnya. Tikus diterapi dengan ekstrak polifenol rumput laut coklat dengan dosis 100 mg/kg BB selama 7 hari berturut-turut. Hasil penelitian menunjukkan bahwa hasil uji fitokimia senyawa ekstrak rumput laut coklat ( Sargassum duplicatum Bory) mengandung senyawa polifenol (flavonoid dan florotanin) dan alkaloid. Hasil KLT preparatif dan spektrum IR noda ekstrak yang terpisah menunjukkan adanya gugus O-H, ikatan C=C aromatik, C-H aromatik, C=C alkena, C-O dan C-H alifatik. Hasil penelitian menunjukkan bahwa terdapat penurunan kadar MDA dan aktivitas protease berturut-turut sebesar 54,20% dan 44,09%. Selain itu, terdapat perbaikan ekspresi protein ZO-1 dan occludin serta perbaikan kerusakan jaringan jejunum usus halus tikus putih berdasarkan pengamatan histologis. Hasil analisis statistik kadar MDA dan aktivitas protease menunjukkan bahwa terdapat perbedaan sangat nyata antara tikus kontrol negatif, tikus sakit yang diinduksi indometasin (IBD) dan tikus yang diterapi ekstrak polifenol rumput laut coklat ( Sargassum duplicatum Bory) (p 0,05).
English Abstract
IBD is a chronic inflammatory disease of the digestive tract especially in small and large intestines. IBD is caused by immune response disturbance which stimulated by virus, bacteria and xenobiotic. One of which stimulate immune response disturbance in small intestine is indomethacin as NSAID ( non-steroidal anti-inflammatory drug ). Indomethacin can stimulate ROS production so that need antioxidant for IBD therapy. The aims of this research were to describe the potency of brown seaweed ( Sargassum duplicatum Bory) polyphenol extract to decrease malondialdehyde (MDA) level and protease activity, to repair both of ZO-1 and occludin protein expressions and rat small intestine jejunum tissue damage which inducted by indomethacin. This research was divided into two stages. The first stage was extraction of polyphenol compounds from brown seaweed, phytochemistry test, preparative thin layer chromatography (TLC) and analysis of extract compounds with infrared spectrophotometry (IR). The second stage was trial of brown seaweed ( Sargassum duplicatum Bory) polyphenol extract to IBD rat. This study used experimental animals of male white rat ( Rattus norvegicus ) wistar strain. Group of sick rat (IBD) induced by indomethacin at a dose 15 mg/kg body weight once orally and incubated for 7 days. IBD rat was expressed after 24 hours induction of indomethacin. The mucosal damage (villous) was based on observation of the small intestine jejunum histological picture. The rat was treated with brown seaweed polyphenol extract with a dose 100 mg/kg for 7 days consecutively. Based on phytochemistry test, brown seaweed extract ( Sargassum duplicatum Bory) contains polyphenol compounds (flavonoids and florotanin) and alkaloid. The result of preparative TLC and IR spectrum of extract spot showed OH group, C=C aromatic, CH aromatic, C=C alkene, CO and CH aliphatic bonds. MDA level and protease activity were respectively decreased, 54.20% and 44.09%. In addition, there were retrieval of ZO-1 and occludin protein expressions as well as retrieval of small intestine jejunum tissue damage of rat by histological observation. Statistical analysis indicated that MDA level and protease activity were extremely difference (p 0.05) on negative control rat, indomethacin-induced (IBD) rat and therapy rat with brown seaweed ( Sargassum duplicatum Bory) polyphenol extract.
