Abstract

Xilanase adalah enzim yang mengkatalisis reaksi hidrolisis xilan menjadi xilosa. Amobilisasi Xilanase digunakan untuk meningkatkan stabilitas dari pengaruh lingkungan, seperti pengaruh protease. Tujuan dari penelitian ini adalah mengkarakterisasi xilanase hasil isolasi Trichoderma viride yang diamobilkan pada matriks zeolit teraktivasi asam. Karakterisasi xilanase amobil yang dipelajari meliputi penentuan kondisi kerja optimum xilanase (pH, temperatur dan waktu inkubasi), nilai kinetik (KM dan Vm), dan efisiensi penggunaan ulang xilanase amobil. Kondisi kerja optimum ditentukan dengan cara mengukur aktivitas pada variasi pH (3, 4, 5, 6, dan 7), temperatur (40, 50, 60, 70, 80) oC, waktu inkubasi (35, 40, 45, 50, dan 55) menit, dan nilai kinetik xilanase ditentukan pada variasi konsentrasi substrat (0,1; 0,5; 1,0; 1,5; 2,0)%. Hasil penelitian menunjukkan bahwa kondisi optimum xilanase amobil dicapai pada pH 6, temperatur 50 °C dan waktu inkubasi 45 menit. Nilai kinetik xilanase amobil diperoleh Vm (29,24 μg.g-1.menit-1) dan KM (1,281%). Xilanase amobil dapat digunakan kembali sebanyak 3 kali dengan efisiensi enzim sisa sebesar 85,2% dan aktivitas enzim sebesar 122,44 μg.g-1.menit-1.

English Abstract

Xylanase is the enzyme that catalyzes the hydrolysis reaction of xylan to be xylosa. Immobilized xylanase was used in order to increase the stability from environmental influences, such as protease influences. The purpose of this research was characterized the xylanase isolated form Trichoderma viride which immobilized in activated zeolit by acid. The characterization of the immobilized xylanase that was studied include the optimum condition of xylanase (pH, temperature and incubation time), kinetic value (KM and Vm), and efficiency of reuse immobilized xylanase. The optimum condition determined by measuring the activity were pH (3, 4, 5, 6, and 7), temperatur (40, 50, 60, 70, 80) oC, incubation time (35, 40, 45, 50, and 55) minutes, and the xylanase kinetic value was studied in various substrate concentration (0.1; 0.5; 1.0; 1.5; 2.0)%. The results showed that the optimum condition of immobilized xylanase were reached at pH 6, a temperature of 50 °C and 45 minutes of incubation time. The kinetic value of immobilized xylanase were Vm (29,24 μg.g-1.minutes-1) and KM (1,281 %). Immobilized xylanase can be reused 3 times with efficiency of 85,2%, enzyme activity remained 122,44 μg.g-1 minutes-1.
