Abstract
Minyak gandapura merupakan minyak atsiri dengan komponen utama metil salisilat. Metil salisilat di dalam daun gandapura dilindungi oleh xilan. Penambahan xilanase untuk menghidrolisis xilan menjadi xilosa sehingga penambahan xilanase dapat mempermudah isolasi minyak gandapura. Penelitian ini bertujuan mempelajari pengaruh penambahan xilanase terhadap rendemen dan kualitas minyak gandapura hasil isolasi dari daun gandapura meliputi destilasi minyak gandapura, uji fisik dan identifikasi komponen penyusun minyak gandapura. Pengaruh penambahan xilanase yang dipelajari meliputi destilasi daun gandapura tanpa penambahan xilanase dan destilasi dengan penambahan xilanase. Uji fisik dilakukan dengan mengukur indeks bias minyak gandapura dengan refraktometer, uji fisik warna dan bau dilakukan pengamatan dengan organoleptik, menentukan kelarutan dengan menggunakana pelarut N-heksana. Identifikasi komponen penyusun minyak gandapura dengan GC-MS. Hasil penelitian menunjukkan rendemen minyak gandapura tanpa penambahan xilanase sebesar 1.42 % sedangkan minyak gandapura dengan penambahan xilanase sebesar 3.34 %. Berdasarkan uji fisik didapatkan indeks bias minyak gandapura tanpa penambahan xilanase sebesar 1,3324 dan indeks bias minyak gandapura penambahan xilanase sebesar 1,5194, tidak berwarna, memiliki bau tajam, larut dalam N-heksana. Hasil identifikasi minyak gandapura tanpa penambahan xilanase didapatkan peak metil salisilat dengan nilai m/z 152 dan peak etil heksanol dengan nilai m/z 57 sedangkan minyak gandapura dengan penambahan xilanase didapatkan peak tunggal metil salisilat dengan nilai m/z 152.
English Abstract
Wintergreen oil is an essential oil with the main component of methyl salicylate. Methyl salicylate in the wintergreen leaves protected by xylan. Addition of xylanase is used for hydrolyzing xylan to xylose in order to isolate wintergreen oil easily. This research has a purpose to study the effect of xylanase addition towards the yield and quality of wintergreen oil isolated from wintergreen leaves including distillation of wintergreen oil, physical tests and identification of components of wintergreen oil. The effects of xylanase addition was a distillation of wintergreen leaves without addition of xylanase and distillation with the addition of xylanase. Physical tests of wintergreen oil conducted by measuring the refractive index with a refractometer, physical tests of colour and odour were observed organoleptically, determined the solubility with n-hexane solvent. Identification the components of wintergreen oil with GC-MS. The results showed that yield of wintergreen oil without addition of xylanase was 1.42% while wintergreen oil with the addition of xylanase was 3.34%. Based on the physical test obtained refractive index of wintergreen oil with addition of xylanase was 1.5194 and refractive index of wintergreen oil without addition of xylanase was 1.3324, colourless, has a pungent odour, soluble in N-hexane. Identification results of wintergreen oil without addition of xylanase obtained methyl salicylate peak with the value of m / z 152 and peak of ethyl hexanol with a value of m / z 57 while wintergreen oil with the addition of xylanase obtained single peak of methyl salicylate with the values m / z is 152.
