Abstract
Pada penelitian ini xilanase diamobilisasi dengan metode adsorpsi fisik menggunakan kaolin. Amobilisasi perlu dilakukan untuk meningkatkan efisiensi pemakaian enzim sehingga dapat dipakai berulang kali. Penelitian ini bertujuan untuk mengetahui waktu pengocokan, konsentrasi xilanase optimum, serta efisiensi pemakaian ulang xilanase amobil. Pada penelitian ini dilakukan variasi waktu pengocokan (1, 2, 3, 4, 5) jam dan konsentrasi enzim sebesar (0,157; 0,209; 0,261; 0,314; 0,366) mg/mL. Kadar protein enzim diuji secara spektrofotometri dengan reagen Biuret dan aktivitas xilanase ditentukan dengan mengukur kadar xilosa secara spektrofotometri menggunakan reagen DNS. Kadar protein xilanase bebas diperoleh sebesar 0,366 mg/mL dengan aktivitas sebesar 8,449 unit. Hasil penelitian menunjukkan bahwa kondisi optimum amobilisasi xilanase pada kaolin dicapai pada waktu pengocokan 3 jam dengan jumlah xilanase teradsorpsi 474 mg/g dan aktivitas 10,074 unit. Sedangkan pada variasi konsentrasi didapatkan konsentrasi xilanase 0,261 ppm dan aktivitas 14,336 unit. Xilanase amobil ini secara efisien dapat digunakan hingga 5 kali pemakaian dengan aktivitas sebesar 51,40%.
English Abstract
In this researh xylanase was immobilized with adsorption method by using kaolin. Immobilization is conducted in order to increase efficiency of enzyme utilization so that it can be used repeatedly. The objective of this research was to determine agitation time, optimum xilanase concentration, and reuse efficiency of immobilized xylanase. This research consisted two treatments which agitation time of (1, 2, 3, 4, 5) hours and enzyme concentrations of (0.157 0.209, 0.261, 0.314, 0.366) mg/mL. Enzyme concentration was analyzed by spectrophotometry with Biuret reagent. Xylanase activity was determined by measuring xylose content with spectrophotometry by using DNS reagent. Free xylanase was 0.366 mg/mL with activity 8.449 unit. The result showed that the optimum condition of immobilized xylanase on kaolin was obtained at 3 hours agitation with total adsorbed xylanase 4,74 mg/g resalting in xylanase actitity of 10.074 unit. Whereas the optimum xylanase concentration was 0.261 ppm yielding in 14.336 unit activity. This Immobilized xylanase can be used 5 times with remained xylanase activity of 51.40%.
