Abstract

Rasio albumin-kreatinin (ACR) merupakan metode pengukuran albumin dan kreatinin pada urin sebagai indikator adanya penyakit albuminuria. Penentuan albumin didasarkan pada metode dye-binding dengan zat warna berupa metil orange sedangkan pada penentuan kreatinin didasarkan pada reaksi Jaffe yaitu reaksi antara kreatinin dengan asam pikrat dalam suasana alkali. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi dan volume metil orange, konsentrasi asam pikrat, volume kreatinin standar/sampel, konsentrasi asam pikrat, volume kreatinin standar/sampel, dan waktu reaksi terhadap absorbansi serta menentukan ACR dalam sampel urin. Penentuan albumin dan kreatinin diukur dengan metode sequential injection analysis (SIA) dengan konsep lab at valve yang baru dikembangkan dengan menambahkan suatu valve mixing untuk memaksimalkan reaksi antara sampel dan reagen. Hasil penelitian menunjukkan kondisi optimum pada penentuan albumin yaitu konsentrasi metil orange 0,005%, volume metil orange 75 μL, dan waktu reaksi 0 detik. Sedangkan kondisi optimum pada penentuan kreatinin yaitu konsentrasi asam pikrat 0,015 M, volume kreatinin standar/sampel 15 μL, dan waktu reaksi 0 detik. English Abstract

Albumin-creatinine ratio (ACR) is a method of albumin and creatinine measurement in urine as an indicator of albuminuria. Determination of albumin is based on dye-binding method with methyl orange while the determination of creatinine based on the Jaffe’s reaction between creatinine and picric acid in alkaline medium. This study aimed to determine the effect of concentration and volume of methyl orange, the concentration of picric acid, standard creatinine/sample volume, and the reaction time of the absorbance and also determine the ACR in the urine sample. Determination of albumin and creatinine were measured by the method of sequential injection analysis (SIA) with the lab at valve concept newly developed valve by adding a mixing valve to maximize the reaction between sample and reagents. The results showed the optimum conditions for the determination of albumin is 0,005% for concentration of methyl orange, methyl orange volume of 75 μL, and the reaction time of 0 seconds. While the optimum conditions in creatinine determination is 0,015 M for concentration of picric acid, standard creatinine/sample volume of 15 μL, and the reaction time of 0 seconds. The detection limit of albumin and creatinine respectively is 0,3521 mg/L and 3,3333 mg/L. This method was applied to the two urine samples with ACR value of each 29,4452 mg / L and 28,1591 mg / L.
