Abstract

Inflammatory Bowel Disease (IBD) adalah penyakit peradangan yang menyerang sistem pencernaan dimana umumnya disebabkan oleh infeksi virus maupun bakteri dan efek samping penggunaan obat anti-inflamasi non-steroid (NSAID) yaitu indometasin. Kerang mas ngur digunakan karena kandungan flavonoid berupa apigenin diharapkan mampu menghambat inflamasi dan memicu perbaikan histopatologi lambung. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak metanol kerang mas ngur terhadap aktivitas protease dan gambaran histopatologi lambung tikus yang telah diinduksi indometasin dengan dosis 15 mg/kgBB selama 1 hari. Pada penelitian ini digunakan hewan coba tikus (Rattus novergicus) yang dibagi dalam 4 kelompok dengan masing-masing kelompok terdiri dari 5 tikus. Empat kelompok tersebut terdiri dari tikus kontrol negatif, tikus kontrol positif (diinduksi indometasin), tikus kelompok induksi indometasin terapi ekstrak metanol kerang mas ngur 400 mg/kgBB, dan tikus kelompok induksi indometasin terapi ekstrak metanol kerang mas ngur 700 mg/kgBB. Aktivitas protease dalam penelitian ini diukur dengan mengunakan metode spektrofotometri UV-Vis sedangkan gambaran histopatologi jaringan lambung menggunakan metode pewarnaan Hematoxylen-Eosin (HE).Hasil penelitian menunjukkan pemberian terapi ekstrak kerang mas ngur mampu mengurangi aktivitas protease pada tikus terapi 400 mg/kgBB dan pada tikus terapi 700 mg/kgBB berbeda sangat nyata (p<0,01) dibandingkan pada tikus IBD. Untuk gambaran histopatologi diketahui bahwa terjadi perbaikan jaringan mukosa lambung pada kelompok tikus terapi.

English Abstract

Inflammatory Bowel Disease (IBD) is an inflammatory disease that attacks the digestive system which is usually cause by a viral or bacterial infection and the side effect of anti-inflammatory drugs non-steroid (NSAID) that is indomethacin. Mas ngur shellfish used because contained flavonoids in this case is apigenin that expected to inhibit inflammation and trigger repair gastric histopathology. This study aims to determine the effect of the methanol extract of mas ngur shellfish against protease activity and gastric histopathology description of mice that had been induced by indomethacin at a dose of 15 mg/KgBW for 1 day. In this study used mice (Rattus novergicus) were devided into four groups whit each group consisting of 5 mice. The four groups consisted of negative control mice, positive control mice (induced by indomethacin), a group of mice induced by indomethacin therapy methanol extract of mas ngur shellfish 400 mg/KgBB, and a group of mice induced by indomethacin therapy methanol extract of mas ngur shellfish 700 mg/KgBW. Protease activity in this study was measured by using UV-Vis spectrophotometry method while histopathology picture of gastric tissue using staining method Hematoxylen-Eosin (HE). The results showed therapy mas ngur shellfish extract can reduce protease activity in mice of therapy of 400 mg/kgBW in mice therapy and 700 mg/kgBW significantly different (p <0.01) compared to IBD mice. For a description of histopathology known that gastric mucosal tissue repair occurs in groups of mice therapy.
