Abstract

Uwi ungu (Dioscorea alata L.) merupakan famili Dioscoreaceae yang mengandung saponin (diosgenin) yang diketahui bermanfaat sebagai agen imunomodulator. Tujuan dari penelitian ini adalah untuk mengetahui adanya perubahan kuantitas sel T CD4+, D8+, CD4+CD62L+, CD8+CD62L+ dan sel B220+IgE+ pada organ lymph node mencit BALB/c model alergi pencernaan pasca pemberian ekstrak etanol umbi uwi ungu, dosis optimum ekstrak etanol umbi uwi ungu dan histolopatologis hepar pasca pemberian ekstrak etanol umbi uwi ungu. Pemberian ekstrak etanol umbi uwi ungu dilakukan pada hari ke 1-28. Hari ke-15 dilakukan injeksi valbumin (OVA) secara intraperitoneal. Kemudian 3 hari setelah injeksi OVA mencit dibedah. Hari ke-22 dilakukan challenge OVA secara intraperitoneal. Hari ke-23 sampai 28 mencit dilakukan injeksi OVA kembali secara oral. Kemudian dilakukan pembedahan pada hari ke-29. Limfosit diisolasi pada organ lymph node dan dilihat profil sel T CD4+, CD8+, CD4+CD62L+, CD8+CD62L+ dan sel B220+IgE+ melalui analisis flow cytometry. Hepar diisolasi dan dibuat preparat histopatologi. Data dianalisis menggunakan One-way ANOVA (p<0,05) dan uji Tukey menggunakan SPSS 16 for Windows. Ekstrak etanol umbi uwi ungu memicu aktivitas imunokompeten mencit model alergi pencernaan. Hal ini diketahui melalui peningkatan jumlah sel T CD4+CD62L+ sebesar 85,29 % dan sel T CD8+CD62L+ sebesar 67,04 % (dosis 0,167 g/kg BB) setelah sensitisasi OVA. Jumlah sel T CD4+, CD8+ dan sel B220+IgE+ menurun signifikan pada dosis 0,167 g/kg BB. Variasi dosis ekstrak etanol umbi uwi ungu berbeda signifikan dan dosis optimum untuk menyembuhkan alergi pencernaan adalah 0,167 g/kg BB. Ekstrak etanol umbi uwi ungu mengurangi nukleus pyknosis pada sel hepatosit dan infiltrasi sel mononuklear pada struktur jaringan hati.

English Abstract

Purple yam tuber (Dioscorea alata L.) is a family Dioscoreaceae that contain diosgenin which is known to be an immunomodulatory agent. The purpose of this study is to understand about the quantitative changes of T cells CD4+, CD8+, CD4+CD62L+, CD8+CD62L+, and B220+IgE+ cells in lymphnode BALB/c mice digestive allergy model after ethanol extract in purple yam tuber oral treatment, the optimum dose of purple yam tuber ethanol extract and the hepar histopatology in BALB/c mice model of digestive allergy after ethanol extract of purple yam tuber treatment. The ethanol extract of purple yam tuber treatment conducted on 1st until 28th days. Then, on 15th day is conducted OVA injection intraperitoneally. After 3 days injection of OVA, 3 mice for each treatment were sectioned. OVA challenge injection intraperitoneally conducted on 22th days. OVA is injected orally on 23th until 28th days. Then, the mice were sectioned on 29th day. Lymphocytes was isolated from lymphnode organs and the profiles of T cells CD4+, CD8+, CD4+CD62L+, CD8+CD62L+, and B220+IgE+ cells were observed through flowcytometry analysis. The hepar was isolated to make the histopathology analysis. Data were analyzed using One-way ANOVA (p <0.05) and Tukey test using SPSS 16 for Windows. Ethanol extract of purple yam tuber triggers the activity immunocompetent T cells in mice model digestive allergy. The result indicated an increasing number of CD4+CD62L+ and CD8+CD62L+ in dose 0.167 g/kg BW. Ethanol extract of purple yam tuber reduced B220+IgE+ in dose 0.167 g/kg BW. Dose variations in extract ethanol of purple yam tuber are significant and the optimum dose of purple yam ethanol extract for digestive allergy was 0.167 /kg BW. Ethanol extract of purple yam tuber reduced the nucleus pyknotic in hepatocytes cells and mononuclear cell infiltration in the liver tissue structure.
