Pengaruh Pupuk Nitrogen terhadap Ukuran dan Kerapatan Kristal Kalsium Oksalat pada Umbi Porang (Amorphophallus muelleri Blume)

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pupuk nitrogen terhadap bentuk, ukuran, dan kerapatan kristal kalsium oksalat pada umbi porang serta berat umbi porang. Bulbil yang berasal dari periode tumbuh pertama disemaikan hingga daun membuka penuh. Tanaman dengan tinggi sekitar 50 cm dipindahkan ke dalam polibeg. Setelah beradaptasi selama tiga minggu, tanaman diberi perlakuan pupuk nitrogen (urea) dengan dosis 0, 1, dan 5 g/polibeg. Tanaman dipelihara hingga akhir periode tumbuh. Preparat dibuat dengan mengiris umbi setipis mungkin menggunakan mikrotom geser. Irisan umbi porang dijernihkan menggunakan metode clearing yang telah dimodifikasi. Preparat diamati menggunakan mikroskop cahaya untuk menghitung kerapatan kristal CaOx serta mengukur panjang dan diameter kristal CaOx. Data ukuran dan kerapatan kristal CaOx dianalisis dengan ANOVA yang dilanjutkan dengan uji Tukey (α = 0,05) jika terdapat perbedaan yang signifikan. Hasil penelitian menunjukkan bahwa umbi porang memiliki empat jenis kristal, yaitu rafida, druse, stiloid, dan prisma. Pemupukan nitrogen tidak berpengaruh terhadap ukuran kristal rafida berkas golongan panjang, rafida tunggal (panjang dan pendek), dan stiloid. Kerapatan kristal kalsium oksalat dan berat umbi porang juga tidak dipengaruhi oleh pemupukan nitrogen. Sebaliknya, pemupukan nitrogen berpengaruh secara nyata terhadap ukuran kristal rafida berkas golongan pendek, druse solid, dan druse tidak solid.

English Abstract

The aim of this research was to know the effects of nitrogen fertilizer to the shape, size, and density of calcium oxalate crystals in tuber of porang and tuber weight of porang. Bulbil derived from the first growth period seeded until the leaves fully open. The plants with a height of about 50 cm were transferred into a polybag. After adapting for three weeks, the plants were treated with nitrogen fertilizer (urea) with a dose 0, 1, and 5 g/polybag. The plants were grown until the end of growth period. The slide were prepared by slicing tuber as thin as possible using a sliding microtome. Sliced tuber of porang were cleared by using the modified clearing method. The slide were observed using light microscope to calculate the density of CaOx crystals and measure the length and the diameter of CaOx crystals. The size of CaOx crystals, densities of CaOx crystals, and tuber weight of porang were analyzed using ANOVA followed by Tukey test (α = 0.05) if there were significant differences. The results showed that tuber of porang has four kind of crystals i.e. raphide, druse, stiloid, and prism. Nitrogen fertilization had no significant effect on the size of bundle raphide crystal long type, single raphide crystal (long and short type), and stiloid crystal. Densities of calcium oxalate crystals and tuber weight of porang also were not affected by nitrogen fertilization. In contrast, nitrogen fertilization significantly affected on the size of bundle raphide crystal short type, solid druse, and unsolid druse.
