Abstract
Penelitian Ini Bertujuan Untuk Mendapatkan Metode Regenerasi Tunas Dari Eksplan Kalus Bawang Putih Dan Menginduksi Tanaman Bawang Putih Tetraploid Secara In Vitro Dengan Kolkisin. Induksi Kalus Dilakukan Dengan Cara Eksplan Meristem Basal Bawang Putih Dikulturkan Pada Medium MS Dengan Penambahan Kombinasi ZPT 2,4-D Dan Kinetin, Sedangkan Regenerasi Tunas Dilakukan Dengan Cara Eksplan Kalus Dikulturkan Pada Medium MS Dengan Penambahan Kombinasi ZPT NAA Dan BAP. Induksi Bawang Putih Tetraploid Dilakukan Dengan Cara Kalus Dikulturkan Pada Medium MS Dengan Penambahan Kolkisin Selama 2 Dan 4 Minggu. Kalus Hasil Perlakuan Kolkisin Disubkultur Pada Medium MS Dengan Penambahan Kombinasi ZPT NAA Dan BAP Untuk Regenerasi Tunas. Level Ploidi Ditentukan Melalui Pengamatan Kromosom Sel Akar Planlet Hasil Regenerasi Dari Perlakuan Kolkisin. Hasil Penelitian Menunjukkan Bahwa Pertumbuhan Kalus Terbaik Terdapat Pada Media MS + 2,4-D 0.3 Ppm + Kinetin 0.5 Ppm. Penambahan Kombinasi NAA 0.1 Ppm Dan BAP 2 Ppm Pada Medium Kultur Mampu Menghasilkan Mata-Mata Tunas, Sedangkan BAP 10 Ppm Mampu Menginduksi Pemanjangan Tunas. Penambahan Kolkisin Pada Medium Kultur Kalus Mampu Menginduksi Poliploidi Bawang Putih, Namun Kolkisin Pada Konsentrasi Tinggi Menghambat Regenerasi Tunas Bawang Putih. Planlet Bawang Putih Tetraploid (13%) Diperoleh Dari Perlakuan Kolkisin 120 Ppm Pada Medium Kultur Kalus Selama 2 Minggu. Selain Itu, Kolkisin Mampu Menginduksi Triploid Dan Mixoploid Pada Planlet Hasil Regenerasi Tunas Bawang Putih.
English Abstract
The Objective Of This Research Were To Get The Plant Regeneration Method From Callus Explants In Garlic And In Vitro Induce The Tetraploid Garlic Using Colchicine. Callus Induction Was Conducted By Culturing Basal Meristem Explants In MS Medium Containing Combination Of ZPT 2,4-D And Kinetin Whereas Shoot Regeneration Was Conduced By Culturing Callus Explants In MS Medium Containing Combination Of ZPT NAA And BAP. Induction Of Tetraploid Was Conducted By Culturing Callus In MS Medium Containing Colchicine For 2 And 4 Weeks. Callus From Colchicine Treatment Were Subcultured In MS Medium By Adding Combination Of ZPT NAA And BAP For Shoot Regeneration. Ploidy Level Was Determined By Observation Cell Chromosome Of The Root From Plantlets Regenerated From Colchicine Treatement. The Observation Results Showed That The Perfect Callus Growth In MS Medium Containing 0.3 Ppm 2,4-D And 0.5 Ppm Kinetin. The Addition Of 0.1 Ppm NAA And 2 Ppm BAP In The Culture Medium Capable Of Induced Small Shoots And 10 Ppm BAP Was Able To Shoot Elongation. The Addition Colchicine In Culture Medium Ofcallus Could Induce The Polyploidy Garlic. However High Consentration Of Colchicine Inhibited The Growth Of Shoot Regeneration In Garlic. Tatraploid Garlic (13%) Were Discovered On 120 Ppm Colchicine Treatement In Culture Medium Of Callus For 2 Weeks. Beside That, Colchicine Was Able To Induce Triploid And Mixoploid Garlic For Planlet That Regenerated From Garlic Shoots.
