Abstract
Tujuan penelitian ini adalah untuk mengetahui efek lama pembekuan semen terhadap kadar MDA spermatozoa dan seminal plasma dan persentase viabilitas spermatozoa. Sampel yang digunakan berupa semen beku sapi Limousin dengan lama pembekuan 2 bulan dan 3 bulan yang dikoleksi dari BBIB Singosari. Semen beku yang telah di thawing diencerkan dengan Phosphate Buffer Saline (PBS) kemudian disentrifugasi untuk memisahkan spermatozoa dan seminal plasma. Kadar MDA spermatozoa dan seminal plasma diukur dengan menggunakan spektrofotometer. Pengamatan viabilitas spermatozoa dilakukan dengan menggunakan pewarnaan eosin-negrosin. Data dianalisis dengan uji-t berpasangan dan korelasi pearson menggunakan spss 16.0. Hasil analisis menunjukkan bahwa kadar MDA spermatozoa pada lama pembekuan 2 bulan (0,0777±0,008 nM) dan 3 bulan (0,0919±0,016 nM) tidak berbeda nyata (p<0,05). Kadar MDA seminal plasma pembekuan 2 bulan (0,1168±0,018 nM) dibandingkan pembekuan 3 bulan (0,1172±0,026 nM) tidak berbeda nyata. Terdapat korelasi negatif antara persentase viabilitas dan kadar MDA baik pada spermatozoa dan seminal plasma.
English Abstract
The aim of this research was to determine the effect of long-term freezing on spermatozoa and seminal plasma MDA levels, and spermatozoa viability. Frozen semen of cattle limousine that had been stored for 2 and 3 months were collected from BBIB Singosari used in this study. The frozen-thawed semen was diluted with Phosphate Buffer Saline (PBS) and then centrifuged to separate the spermatozoa and seminal plasma. The MDA levels of spermatozoa and seminal plasma were measured by spectrophotometer. Spermatozoa viability was assessed using Eosin-Negrosin staining. The data were analyzed using paired t-test and Pearson correlation employing SPSS 16.0 for Windows. The result showed that MDA levels of spermatozoa for 2 months storage (0.0777 ± 0.008 nM) and 3 months storage (0.0919 ± 0.016 nM) were not significant differences (p<0.05). Seminal plasma MDA levels were not significant differences between 2 months storage (0.1168 ± 0.018 nM) and 3 months storage (0.1172 ± 0.026 nM). There is negative correlation between the percentage of viability and both spermatozoa and seminal plasma of MDA levels
