Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun tempuyung (Sonchus arvensis) terhadap jumlah sel yang mengekspresikan CuZnSOD pada ginjal mencit model nefrolithiasis. Mencit (Mus musculus) jantan galur Swiss berusia delapan minggu dibagi menjadi kelompok (A) kontrol, (B) nefrolithiasis, (C) ekstrak daun tempuyung, (D) nefrolithiasis dan ekstrak daun tempuyung, dan (E) nefrolithiasis dan pemberian ekstrak daun tempuyung secara simultan. Untuk membuat model nefrolithiasis dilakukan dengan pemberian tepung umbi porang (Amorphophallus muelleri) secara oral dengan dosis 0,06 mg/g BB setiap hari selama tiga bulan. Ekstrak daun tempuyung diperoleh dari produk CalcusolTM yang diproduksi Perusahaan Obat Tradisional dr. Sardjito, Yogyakarta. Dosis calcusol yang diberikan adalah 3,3 mg/g BB. Setelah perlakuan, mencit didislokasi leher, dibedah dan diisolasi ginjal untuk pembuatan preparat parafin. Analisis CuZnSOD menggunakan teknik immunohistokimia, menggunakan antibodi primer poliklonal rabbit anti-SOD1 (Bioss, bs-1079R). Preparat diamati dengan mikroskop Olympus BX51 perbesaran 400x. Gambar didokumentasikan dengan Olympus Digital Camera DP20. Persentase sel yang mengekspresikan CuZnSOD dianalisis menggunakan perangkat lunak Immunoratio (URL:http://153.1.200.58 :8080/immunoratio/). Hasil menunjukkan pemberian ekstrak daun tempuyung dapat menurunkan jumlah sel yang mengekspresikan CuZnSOD dalam organ ginjal model mencit nefrolithiasis secara signifikan (P<0,05). Diduga senyawa antioksidan dalam ekstrak tempuyung mampu mereduksi ROS secara langsung tanpa harus menginduksi sel untuk memproduksi enzim CuZnSOD.
English Abstract
This study was to investigate the effect of tempuyung leaves (Sonchus arvensis) extract on number of cells expressing CuZnSOD in mice renal of nephrolithiasis model. Eight weeks old Swiss strain male mice (Mus musculus) were divided into five groups: (A) control, (B) nephrolithiasis, (C) tempuyung leaves extract treatment, (D) nephrolithiasis & tempuyung leaves extract treatment, and (E) nephrolithiasis & tempuyung leaves extract treatment simultaneously. Nephrolithiasis was induced orally by applying porang tuber (Amorphophallus muelleri) flour 0,06 mg/g BW for three months daily. Tempuyung extract was obtained from CalcusolTM product by Traditional Medicine Company dr. Sardjito, Yogyakarta. Calcusol dosage administered to mice was 3,3 mg/g BW. After three months, mice were killed by neck-dislocation, the kidneys were isolated and prepared for paraffin histology. CuZnSOD was analyzed by immunohistochemistry (IHC), using rabbit policlonal antibody anti-SOD1 (Bioss, bs-1079R) as primary antibody. Tissues were observed under Olympus BX51, 400x magnification. Images were documented with Olympus Digital Camera DP20. The percentage of number of cells expressing CuZnSOD were analized in Immunoratio software online (URL: http://153.1.200.58:8080 /immunoratio/). The result showed that S. arvensis extract administration was able to decrease number of cells expressing CuZnSOD in kidney significantly (P<0.05). It was supposed that antioxidant content in S. arvensis leaf enable to scavenge ROS directly with no induction of CuZnSOD production in cell.
