Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh pengencer susu kedelai bubuk berdasarkan konsentrasinya F1(2%), F2 (4%) dan F3 (6%) dalam pengencer sitrat terhadap motilitas, viabilitas, abnormalitas dan integritas membran spermatozoa berdasarkan lama penyimpanan (0 jam, 24 jam, 72 jam, 120 jam). Penelitian ini menggunakan media pengencer berbasis media nabati (lesitin) yaitu Andromed® (K+), susu kedelai dalam pengencer sitrat dengan konsentrasi 0% (K-), 2% (F1), 4% (F2) dan 6% (F3). Pengamatan dilakukan sebelum pengenceran yaitu pengamatan volume, warna, konsistensi, pH, motilitas massa, motilitas individu, viabilitas, konsentrasi, dan abnormalitas. Selanjutnya dilakukan pengenceran berdasarkan perlakuan dengan masing-masing tiga ulangan dan dilakukan penyimpanan pada suhu 4oC, lalu pengamatan pasca penyimpanan yaitu motilitas, viabilitas, abnormalitas, dan integritas membran. Penelitian ini menggunakan RAL faktorial dan dianalisis menggunakan two way ANOVA dengan program SPSS versi 16.0 dan dilanjutkan dengan uji Tukey. Hasil penelitian secara umum menunjukkan adanya beda nyata (P<0,05) untuk pengencer K+, F1, dan F2 terhadap pengencer K- dan F3. Adanya kandungan lesitin pada susu kedelai mampu melindungi spermatozoa dari cekaman dingin (cold shock) selama penyimpanan pada suhu 4 oC. Konsentrasi susu kedelai bubuk yang terlalu tinggi kemungkinan menyebabkan perubahan pH pada media dan mempengaruhi tekanan osmotik pada membran spermatozoa sehingga menurunkan kualitas spermatozoa selama penyimpanan
English Abstract
The objective of this research was to know the influence of soybean milk powder diluents based on concentration (2%, 4%,6%) in citrate diluent to motility, viability, abnormality and membrane integrity of spermatozoa based on long duration storage (0 hours, 24 hours, 72 hours, 120 hours). This research using diluent medium based on vegetable medium (lechitin) is andromed (K+), soybean milk in diluent citrate with concentrations of 0% (K-), 2% (F1), 4% (F2), and 6% (F3). The characterization of predilution were observed such as volume, color, consistency, pH, mass motility, individual motility, viability, concentration, and abnormality. Next step dilution based on treatment with each of the three recurrence and performed storage at temperature of 4˚C. The next of post storage were observed such as sperm viability, motility, abnormality and membrane integrity. Data were analyzed using two way ANOVA in factorial completely randomized design with SPSS version 16.0 and continued with Tukey test. The results showed that a significant difference (P<0.05) to K+, F1, and F2 diluent to K- and F3 diluent. The presence of lechitin content of soybean milk can protect spermatozoa from cold shock during storage at a temperature of 4˚C. The concentration of soy milk powder high possibility changes to pH of the media and influenced of osmotic presure on sperm membrane decrease of the sperm quality during storage.
