Abstract

Fusarium sp. merupakan salah satu patogen tanaman strawberry yang dapat menurunkan hasil panen tanaman inang sekitar 40 %. Penelitian ini bertujuan untuk mempelajari potensi Genus Trichoderma dalam mengendalikan Fusarium sp. Trichoderma diisolasi dari tanah rhizosfer tanaman strawberry, sedangkan Fusarium sp. diisolasi dari tanaman strawberry yang mengalami layu Fusarium. Isolat kapang dimurnikan, dikarakterisasi dan dibandingkan dengan isolat kapang acuan. Uji Antagonis dilakukan secara in vitro dan in vivo. Uji in vitro dilakukan dengan metode dual culture dan slide culture. Hasil penelitian didapatkan dua jenis kapang antagonis yang mirip dengan Genus Trichoderma dan dua jenis kapang patogen yang mirip dengan Genus Fusarium. Isolat antagonis yang diperoleh adalah Trichoderma sp.1 dan Trichoderma sp. 2, serta isolat patogen Fusarium sp. 1 dan Fusarium sp. 2. Isolat Trichoderma sp. 1 memiliki kemampuan antagonisme lebih tinggi dibandingkan dengan isolat Trichoderma sp. 2. Isolat Trichoderma sp. 1 mampu menghambat pertumbuhan Fusarium sp. 1 dan Fusarium sp. 2 secara berturut- turut 49,7 % dan 49,6 %. Isolat Trichoderma sp. 2 mampu menghambat pertumbuhan Fusarium sp. 1 dan Fusarium sp. 2 secara berturut-turut 45,8 % dan 43,4 %. Mekanisme antagonis yang terjadi antara kapang antagonis dan patogen pada uji in vitro yaitu pembelitan dan intervensi hifa. Perlakuan paling optimal pada uji in vivo adalah Trichoderma yang diinokulasi terlebih dahulu yang dilanjutkan dengan penginokulasian Fusarium dengan nilai intensitas serangan sebesar 41,72 % pada varietas Santung sedangkan varietas California yang relatif lebih rentan terhadap serangan patogen.

English Abstract

Fusarium sp. is one of pathogen in strawberrys plant which can decreas harvest product in host plant around 40%. This research aims to study the potential of Genus Trichoderma in controlling Fusarium sp. Trichoderma was isolated from rhizosfer soil in strawberrys plant, while Fusarium sp. was isolated from strawberrys plant which infected by Fusarium wilt. Fungus isolat were purified, characterized and compared by reference isolat. Antagonist test was treated with in vitro test and in vivo test. In vitro test was treated by dual culture and slide culture. The result of the analysis show that it were founded two species of antagonist fungus having similarity to Trichoderma Genus and two species of pathogen fungus having similarity to Fusarium Genus. Antagonist isolat succesfully isolated are Trichoderma sp. 1 and Trichoderma sp. 2, while pathogen isolat succesfully isolated are Fusarium sp. 1 and Fusarium sp. 2. Trichoderma sp. 1 isolat has higher antagonist ability compared to Trichoderma sp. 2 isolat. Trichoderma sp. 1 isolat can block Fusarium sp. 1 and Fusarium sp. 2 growth serially 49,7 % and 49,6 %. Trichoderma sp. 2 isoat is able to block Fusarium sp. 1 and Fusarium sp. 2 growth serially 45,8 % and 43,4 %. Antagonist mechanisms occured in antagonist and pathogen fungus in in vitro test are twisted and miselium intervention. The most optimal treatment in in vivo test is by spreading Trichoderma, then continued by spreading Fusarium with intensity value 41,72 % in Santung variety whereas California variety which is sensitive to pathogen attack.
