Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh beberapa konsentrasi sukrosa dan glukosa terhadap pertumbuhan dan akumulasi betasianin kalus Celosia argentea pada fase logartimik. Kalus diinduksi dari eksplan kotiledon hasil perkecambahan in vitro pada medium MS dengan ZPT NAA 2 ppm dan BAP 2 ppm. Pengaruh beberapa konsentrasi sukrosa dan glukosa masing-masing 1%, 2%, 3%, 4%, 5%, 6% terhadap pertumbuhan (berat basah dan kering) dan akumulasi betasianin diamati pada kultur kalus selama fase log (6, 12, dan 18 hari). Kalus kering diperoleh dengan menggunakan freeze drying. Betasianin diperoleh dari hasil ekstrasi kalus kering dengan pelarut methanol 80% dan asam askorbat 55 mM kemudian kadarnya diukur dengan spektrofotometer pada λ 536 nm. Pertumbuhan kalus C. argentea berdasarkan berat basah dan berat kering pada fase logaritmik (6, 12 dan 18 hari) tidak dipengaruhi oleh jenis dan konsentrasi sumber karbon tetapi dipengaruhi oleh lama kultur. Pertumbuhan kalus yang optimum dihasilkan 1,744 g dan 1,475 g pada lama kultur 18 hari dibandingkan lama kultur 6 dan 12 hari baik pada perlakuan sukrosa maupun glukosa. Kandungan betasianin pada fase logaritmik (6, 12 dan 18 hari) hanya dipengaruhi oleh konsentrasi sumber karbon serta interaksi antara jenis sumber karbon dan lama kultur. Kandungan betasianin tertinggi (31,2 μg/g berat kering) dihasilkan bila ditambahkan sumber karbon 2%. Kandungan betasianin meningkat secara signifikan pada lama kultur 12 hari dan 18 hari masing-masing pada perlakuan sukrosa dan glukosa.
English Abstract
The aim of reaserch was to know the effect of sucrose and glucose in various concentrations on growth and accumulation betacyanin callus Celosia argentea in logarithmic phase. Callus induced in MS medium supplemented with 2 ppm NAA and 2 ppm BAP from in vitro-derived cotyledone explants the effect of sucrose and glucose in various concentration 1%, 2%, 3%, 4%, 5%, 6% on growth (wet and dry weight) and accumulation betacyanin were observed on callus cutured in logarithmic phase (6, 12 and 18 days). Callus dry weight was obtained by freeze drying. Betacyanin was obtained from ectract of callus dry weight dissolved by methanol 80% and 55 mM ascorbic acid, and then the content was measured by spectrofotometry at λ 536 nm. Growth of C. argentea callus in logarithmic phase (6, 12 and 18 days) was not influenced by kind and concentration of carbon sources but influenced by length of culture. Optimum growth of callus resulted 1,744 g and 1,475 g was obtained at 18 days by addition of either sucrose or glucose. Betacyanin accumulation in logarithmic phase (6, 12 and 18 days) was influenced by concentration of carbon sources and interaction between kind of carbon sources and length of culture. The highest of betacyanin content (31,2 μg/g dry weight) was obtained if the medium was supplemented with carbon source 2%. Betacyanin accumulation increase significantly at 12 days and 18 days of culture supplemented with sucrose and glucose, respectively.
