Abstract
Formalin adalah larutan metanal dalam air yang sangat berbahaya jika terhirup, mengenai kulit dan tertelan. Jika terkonsumsi dalam jangka panjang, formalin dapat merusak organ salah satunya adalah ginjal. Ginjal mempunyai peranan penting dalam tubuh karena mengekskresikan bahan buangan. Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi susu kedelai pada tikus putih yang telah terpapar formalin terhadap pencegahan kerusakan MDA dan gambaran histopatologi jaringan ginjal. Pada penelitian ini, tikus putih dikelompokkan menjadi 5 kelompok, yaitu kelompok kontrol, kelompok yang diberi susu kedelai tanpa terpapar formalin, kelompok terpapar formalin, kelompok yang dipapar formalin dengan dosis 17 ppm sekaligus diterapi dengan susu kedelai (preventif), dan kelompok yang dipapar formalin dengan dosis 17 ppm kemudian diterapi dengan susu kedelai (kuratif) yang dipreparasi selama 12 hari. Pada hari ke-13 tikus dibedah dan ginjal diambil lalu diukur kadar MDA serta perubahan gambaran histopatologi organ menggunakan metode pewarnaan Hematoksilin-Eosin. Pengukuran kadar MDA dilakukan menggunakan metode Thiobarbituric Acid (TBA) dengan spektrofotometri UV-Vis pada panjang gelombang 531,8 nm. Hasil penelitian menunjukkan bahwa kadar MDA berturut-turut berdasarkan kelompok tikus putih adalah (3,927 0,056) g/mL, (3,047 0,043) g/mL, (5,617 0,025) g/mL, (4,312 0,049) g/mL dan (5,079 0,029) g/mL. Susu kedelai dapat menurunkan kadar MDA sebesar 23,23 % untuk preventif dan 9,58 % untuk kuratif. Selain itu, susu kedelai dapat memperbaiki histopatologi jaringan ginjal tikus putih yang rusak akibat paparan formalin
English Abstract
Formaline is methanal solution in water, which can be harmful through inhalation, skin contact, or swallowed. Long period consumption of formaline could damage organs, such as kidney. Kidney has an important role in the body for excreting process. The aim of this study was to determine the effect of soy milk supplementation on rats exposed with formaline to repair MDA damage and histopathological kidney tissue. In this study, white rats were categorized into 5 groups, which were control group, rats without formaline exposure and feed with soy milk group, formaline exposure group, formaline exposure group (preventive) : the group that exposed by formaline with the dosage was 17 ppm at once that treated with soy milk, and formaline exposure group (curative) : the group that exposed by formaline with the dosage was 17 ppm then treated with soy milk that had been prepared for 12 days. On the 13th day, the white rat was dissected and the kidney was taken to measure MDA level and the changing of histopathological description in organ using Haematoxylin-Eosin staining method. The MDA level was measured using Thiobarbituric Acid (TBA) method with UV-Vis spectrophotometry at wave length 531.8 nm. The result showed that MDA levels based on the group of white rats were (3.927 0.056) g/mL, (3.047 0.043) g/mL, (5.617 0.025) g/mL, (4.312 0.049) g/mL and (5.079 0.029) g/mL respectively. The soy milk supplementation can reduced MDA level from 23.23 % to 9.58 % for preventive and curative. In addition, it could repair the histopathology of kidney tissue of rats damaged with formaline exposure.
