Abstract

Penelitian ini bertujuan untuk melakukan sintesis dan karakterisasi senyawa turunan α-pinena yang dihasilkan dari reaksi Ritter antara α-pinena dengan asetonitril, serta pengaruh toksisitas senyawa turunan α-pinena hasil dari sintesis terhadap A. salina melalui uji toksisitas dengan metode Brine Shrimp Lethality Test (BSLT). Senyawa hasil isolasi minyak terpentin dan hasil sintesis melalui reaksi Ritter dilakukan karakterisasi terhadap komponen penyusunnya dengan menggunakan Spektrometer Infra Merah dan Kromatografi Gas-Spektrometri Massa (KG-SM), kemudian dilakukan uji toksisitas terhadap A.salina dengan menggunakan metode BSLT. Minyak terpentin diisolasi melalui distilasi fraksinasi dengan pengurangan tekanan yang menghasilkan distilat berupa α-Pinena sebesar 91,33%. Reaksi Ritter antara α-Pinena dengan asetonitril dan H2SO4 menghasilkan senyawa yang bervariasi berdasarkan kestabilan dan penataan ulang karbokation, yaitu N-[1-metil-1-(4-metil-sikloheks-3-enil)-etil]-asetamida (51,31%) sebagai produk utama serta senyawa N-(2,6,6,-trimetilbisiklo(3.1.1) hept-2-il) asetamida (4,86%) dan N-(1,5-dimetil-1-vinil-heks-4-enil)-asetamida (20,29%) sebagai produk samping. Uji toksisitas terhadap A. salina menunjukkan bahwa senyawa hasil sintesis melalui reaksi Ritter dengan nilai LC50 sebesar 34,626 ppm berpotensi sebagai antimikroba.

English Abstract

The aims of the research were to synthesize and characterize α-pinene derivative compounds produced through Ritter reaction between α-pinene and acetonitrile, and the influence of toxicity on the product compounds of Ritter reaction towards A. salina through a toxicity test with Brine Shrimp Lethality Test (BSLT) method. The compounds produced from turpentine oil isolation and synthesis through Ritter reaction is characterized by Infra Red Spectrophotometer and Gas Chromatography-Mass Spectrometry (GC-MS), which is then continued by conducting toxicity test towards A. salina by the BSLT method. Turpentine Oil is isolated through fractional distillation under diminished pressure that produced 91,33% of α-pinene as the distillate. Ritter Reaction between α-Pinene with acetonitrile and H2SO4 resulted in varieties of products based on its carbocation stability and rearrangement, which are N-[1-Methyl-1-(4-methyl-cyclohex-3-enyl)-ethyl]-acetamide (51,31%) as the main product and N-(2,6,6,-trimethylbicyclo(3.1.1) hept-2-yl) acetamide (4,86%) and N-(1,5-Dimethyl-1-vinyl-hex-4-enyl)-acetamide (20,29%) as the byproducts. The toxicity test towards A. salina showed that the products resulted from Ritter reaction with LC50 value of 34.626 ppm has the potential as antimicrobial activity.
