Abstract
Kolam nitrifikasi-denitrifikasi merupakan metode biodegradasi limbah cair yang ada di PT Pupuk Kaltim. Kolam nitrifikasi yang kurang terkontrol faktor biotik dan abiotiknya menyebabkan penurunan kemampuan bakteri nitrifikasi dalam mendegradasi limbah amonium menjadi senyawa nitrit dan nitrat. Penelitian ini bertujuan untuk mengetahui karakteristik dari bakteri pengoksidasi amonium atau Ammonium Oxidizing Bacteria (AOB) dan pengoksidasi nitrit atau Nitrit Oxidizing Bacteria (NOB) serta tingkat toleransinya pada proses nitrifikasi amonium konsentrasi tinggi. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan dua faktor yaitu konsentrasi amonium (500, 600, 700, 800, 900, 1000 mg/l) dan lama waktu inkubasi (24, 48, 72 dan 96 jam) dalam sistem tertutup. Isolat AOB dan NOB dikarakterisasi secara fenotipik dan dianalisis similaritasnya berdasarkan Simple Matching Method . Parameter yang diukur meliputi konsentrasi amonium, nitrit, dan nitrat serta densitas sel bakteri AOB dan NOB. Isolat AOB dan NOB hasil isolasi berada dalam genus yang berbeda dengan nilai similaritas sebesar 56%. Konsentrasi amonium 500, 600, 700, 800, 900 hingga 1000 mg/L merupakan kisaran konsentrasi yang dapat ditoleransi bakteri pengoksidasi amonium (AOB) dan pengoksidasi nitrit (NOB). Konsorsium AOB dan NOB pada inkubasi 48 jam mampu menurunkan amonium hingga 100%, menghasilkan nitrit dan nitrat dengan rata-rata sebesar 95,9% dan 87,31% dalam sistem tertutup skala laboratorium.
English Abstract
The nitrification and denitrification pond is a method of waste liquid biodegradation treatment in PT Pupuk Kaltim. Nitrification pond which uncontrolled of biotic and abiotic factors was caused the failure of nitrifying bacteria to degrade ammonium in waste liquid into nitrite and nitrate. The objective of this research was to study the characteristics of ammonium oxidizing bacteria (AOB) and nitrite oxidizing bacteria (NOB) and the tolerance rates in nitrification of high ammonium concentration treatment. This research was used complete random design with two factor that were ammonium concentration treatment (500, 600, 700, 800, 900, 1000 mg/l) and incubation time (2, 48, 72 and 96 h) in batch culture. The similarity of isolate AOB and NOB were characterisized and analyzed by Simple Matching Method (SS M). The parameters measured were concentration of ammonium, nitrite, nitrate and cell dencities of AOB and NOB. Isolate AOB and NOB were in different genus with similarity percentage is 56%. Ammonium concentration treatment still in tolerance range of AOB and NOB. The consortium of AOB and NOB in 48 h of incubation were able to decrease ammonium concentration as much as 100%, increasing nitrit and nitrat concentration drastically as much as 95,9% and 87,31%.
