Abstract
Salah satu hama utama tanaman jeruk yang menurunkan produktivitas jeruk di Indonesia adalah kutu sisik coklat (Lepidosaphes beckii Newman). Hama tersebut diketahui dapat dikendalikan oleh kapang entomopatogen. Penelitian ini bertujuan untuk mengetahui patogenisitas kapang terhadap kutu sisik coklat ( Lepidoshapes beckii Newman) hama tanaman jeruk dan karakter fenotip kapang entomopatogen isolat Bali. Tahapan penelitian meliputi isolasi kapang entomopatogen dari kutu sisik coklat yang terinfeksi kapang pada buah jeruk, seleksi kapang berdasar patogenisitas, penghitungan viabilitas konidia kapang pada media Potato Dextrose Agar (PDA) dan uji patogenisitas kapang entomopatogen terhadap kutu sisik coklat ( L. beckii ). Selain itu juga dilakukan karakterisasi fenotip kapang (hifa, konidia dan konidiofor) untuk mengetahui nilai similaritas kapang. Hasil isolasi diperoleh 11 isolat kemudian diseleksi menjadi dua isolat terpilih yaitu isolat B 1 B dan B 12 B yang efektif sebagai patogen pada kutu sisik coklat. Isolat B 12 B memiliki kemampuan patogenisitas lebih tinggi daripada isolat B 1 B dengan nilai mortalitas kutu sisik coklat sebesar 48%. Isolat B 1 B memiliki nilai similaritas sebesar 79% dengan isolat PWRJ I D4 ( Fusarium culmorum ) sedangkan isolat B 12 B memiliki nilai similaritas sebesar 74% dengan isolat PC ORG D4 ( Gibberella sp.).
English Abstract
One of the citrus crop pests that decrease of orange productivity in Indonesia was purple scale insect ( Lepidosaphes beckii Newman). The biological control which known to infect and control purple scale insect were entomopathogenic fungus. This aims of this research were to determine the pathogenicity of fungi against purple scale insect ( Lepidoshapes beckii Newman) on citrus plants and phenotypic characters of fungi isolated from Bali. The method used in this research were involved isolation of entomopathogenic fungi from purple scale insect which was infected by fungus in fruit orange. That entomopathogenic fungi was selection based on fungi pathogenicity. Conidia viability was confirmed in Potato Dextrose Agar medium used of 10 conidia/ml. The pathogenicity test of 2entomopathogenic fungi toward purple scale insect ( L. beckii ). Phenotypic characterization of fungi (hyphae, conidia and conidiofor) were conducted to know the similarity of fungi. The result of isolation process were 11 isolates and selected isolates which infected purple scale insect were B 1 B and B 12 B. Isolate B 12 B had 48% mortality value higher than isolate B 1 B had 11,33% mortality value of purple scale insect. Isolate B 1 B had 79% similarity value with isolate PWRJ I D4 ( Fusarium culmorum ) and isolate B 12 B had 74% similarity value with isolate PC ORG D4 ( Gibberella sp.).
