Abstract

Splenectomy is an organ removal of the spleen due to organ function abnormality. In post splenectomy, patients were vulnerable to infection and require compensation for the amount of lymphocytes to handle pathogens that invade the body. IL-2 is one of the compensators agents involved in development of T lymphocytes. The purpose of this experiment are to determine the effect of S. typhi exposure to the levels of IL-2, the relative amount of CD4+ and CD8+ T cells and the effect of S. typhi penetration at small intestine of splenectomized BALB/c mice. This experiment use Complete Random Design with 4 treatment groups (control, splenectomy, S. typhi exposure (109cell/ml), and splenectomy-S. typhi exposure 109cell/ml). Measurement of IL-2 levels in serum of mice performed using ELISA (λ=405 nm), number of CD4+ T cells and CD8+ calculated by flow cytometry and the effects S. typhi penetration in the small intestine of mice was observed with Hematoxylin-Eosin method. The results showed that S. typhi exposure in splenectomized and nonsplenectomized mice can increase levels of IL-2 (control = 0,494 ng/ml, splenectomy-S. typhi exposure = 1,252 ng/ml, S. typhi exposure = 1,352 ng/ml, and splenectomy = 1,567 ng/ml), which indicates the existence of the body’s immune system in the absence of spleen and it’s when exposure by bacteria. The analysis results of the number CD4+ and CD8+ T cells in the mesenteric lymph nodes showed that S. typhi exposure in all treatment mice does not affect on the relative amount of CD4+ and CD8+ T cells. S. typhi penetration in the small intestine of splenectomy-S. typhi exposure mice showed the worst penetration than other groups, indicated by the damage of intestinal tissue structure and the number of bacteria colonies in the small intestine tissue.
