Abstract

Protease merupakan enzim pencernaan yang dapat menghidrolisis protein. Protease banyak diproduksi di saluran cerna seperti jejunum. Kerusakan patologi oleh formalin mengakibatkan penurunan aktivitas protease, yang disebabkan oleh gugus karbonil pada formaldehid yang bersifat sangat reaktif terhadap protein tubuh. Formaldehid dapat mendorong tumbuhnya ROS ( Reactive Oxygen Spesies ) yang menghasilkan senyawa radikal bebas dalam jumlah besar dan bersifat sangat toksik. Pada penelitian ini dipelajari aktivitas protease dari jejunum tikus putih ( Rattus norvegicus ) yang terpapar formalin dan pasca terapi yoghurt. Paparan formalin yang diberikan pada tikus putih sebesar 25 ppm secara oral selama 6 hari kemudian diberi terapi yoghurt sebanyak 2 mL dengan konsentrasi 6,8x10 6CFU/mL secara oral selama 6 hari. Hasil penelitian menunjukkan bahwa pemberian yogurt dapat meningkatkan aktivitas protease sebesar 1,912 % dari 2,088±3,7.10-2 μmol/mL menit menjadi 2,092±4,1.10-2 μmol/mL menit. Perbaikan saluran jejunum pasca terapi yoghurt akan mengakibatkan aktivitas protease meningkat. Peningkatan aktivitas protease menyebabkan berkurangnya sel radang sehingga terjadi perbaikan pada gambaran histologis dari jejunum tikus yang terpapar formalin.

English Abstract

Protease is one of the absorption enzyme which is hydrolyze protein. Protease mainly produced in gastrointestinal track, especially in jejunum. Patology destruction of jejunum that was caused formalin induction cause the decreasing of protease’s activity that is caused by the carbonil cluster in the formaldehid. It is so reactive to body. Formaldehid can support the grow up of ROS ( Reactive Oxygen Spesies which produces deliverance of radical compound in a large number and so toxic. The research learned about the protease activity from Rattus norvegicus jejunum which was induced by formaldehid and after yoghurt therapy. Formalin induced to the group of Rattus norvegicus . The group were induced by 25 ppm orally for 6 days, than was given 2 mL of yoghurt therapy with consentrat of 6,8 x 10 6CFU/mL orally for 6 days. The result experinment that giving the yoghurt can increase the protease activity by 1,912% from 2,088± 3,7.10-2 μmol/mL minute became 2,092±4,1.10-2 minute. Jejunal recovered post yoghurt therapy made an increasing of protease activity. Increasing of protease’s activity caused the decrease of damage cell, so improving the description of histologist from the jejunum Rattus norvegicus that were induced by formaldehid.
