Abstract
Studi terdahulu menguji bahwa leptin menyebabkan terjadinya disfungsi endotel yang merupakan patobiologi aterogenesis. Puerarin merupakan antioksidan yang diduga mampu menghambat mekanisme aterogenesis yang salah satunya dipicu oleh TNF- sebagai zat proinflamasi. Penelitian ini bertujuan untuk menguji dan menentukan berapa konsentrasi puerarin yang dapat menurunkan konsentrasi Tumor Necrosis Factor- (TNF-) pada kultur Human Umbillical Vein Endothelial Cells (HUVECs) yang diinduksi 25 ng/mL leptin. Kultur sel endotel dikelompokkan menjadi empat yaitu kultur sel endotel (1) 0 ng/mL dan 0 M puerarin, (2) diinduksi leptin konsentrasi 25 ng/mL selama 12 jam, (3) diinduksi puerarin masingmasing 5; 25; 200; dan 525 M selama 12 jam tanpa leptin, (4) diinduksi 25 ng/mL leptin bersama puerarin masing-masing 5; 25; 200; dan 525 M selama 12 jam. Selanjutnya dilakukan pengukuran konsentrasi TNF-a menggunakan prosedur dalam human TNF- ELISA Kit pada masing-masing medium kultur sel endotel. TNF-dibaca ELISA reader berdasarkan nilai Optical Density (OD). Konsentrasi TNF- dihitung dengan cara memplotkan nilai OD pada kurva standart TNF-. Data yang diperoleh dianalisis menggunakan SPSS dengan uji ANOVA satu arah dan uji lanjutan Tukey . Hasil penelitian menunjukkan bahwa 25 mM puerarin menurunkan konsentrasi TNF- pada kultur sel endotel vena umbilikus manusia (HUVECS) yang diinduksi leptin 25 ng/mL, sedangkan 200 mM dan 525 mM puerarin menurunkan konsentrasi TNF- mencapai keadaan normal pada kultur HUVECs yang diinduksi leptin 25 /mL.
English Abstract
The last study has proven that leptin caused the endothelial dysfunction as a step of the atherogenesis. Puerarin, an antioxidant, is presumed having the ability to block the atherogenesis mechanism, which is stimulated by TNF- as a proinflamatory cytokine. Therefore, the aim of this research was to prove and to determine whether puerarin could decrease the Tumor Necrosis Factor- (TNF-) concentration in Human Umbillical Vein Endothelial Cells (HUVECs) culture induced by 25 ng/mL leptin. In vitro study used primary culture of the HUVECs were devided in to four groups, there were (1) 0 ng/mL and 0 M puerarin, (2) induced by 25 ng/mL leptin for 12 hours, (3) induced by puerarin each 5; 25; 200 and 525 μM for 12 hours without leptin, (4) induced by leptin and puerarin with concentration 5; 25; 200 and 525 μM for 12 hours. Then the measurement of Tumor Necrosis Factor- (TNF-) concentrations was applied by using human TNF- ELISA Kit procedure on cell endothel culture medium. TNF- was read by ELISA reader based on Optical Density (OD) value. TNF- concentrations were estimated by plotting OD value in standard curve of TNF-. The results were analyzed using SPSS with one-way ANOVA test then followed by Tukey test 0.05. It was showed that puerarin 25 mM decreased TNF- concentration in Human Umbillical Vein Endothelial Cells (HUVECs) culture induced by leptin 25 ng/mL, however puerarin 200 mM and 525 mM decreased TNF- concentration in HUVECs culture induced by leptin 25 ng/mL until reach normallity.
