Abstract
Konsentrasi urea dapat ditentukan secara potensiometri menggunakan biosensor urea. Biosensor urea dibuat dengan melapisi permukaan elektroda gelas menggunakan urease amobil. Kinerja biosensor urea dipengaruhi oleh massa urease amobil. Kinerja biosensor ini ditunjukkan oleh karakteristik biosensor seperti kisaran konsentrasi, batas deteksi, faktor Nersnt, waktu respon dan efisiensi biosensor. Pada penelitian ini, dipelajari optimasi massa urease amobil terhadap karakter biosensor urea pada kisaran massa 0,25g hingga 2,00g. Karakter biosensor dapat diketahui dengan mengukur besarnya potensial sel larutan urea pada kisaran konsentrasi 10-6M hingga 1M. Dari hasil penelitian, diperoleh kinerja biosensor urea terbaik terdapat pada massa urease amobil 1,00g, bilangan Nernst 27,500 mV/dekade. Kisaran konsentrasi urea yang dapat diukur adalah 10-3 M hingga 10-1 M dan batas deteksi sebesar 0,38.10-3 M. Waktu respon biosensor adalah 30 detik dan efisiensi biosensor sampai dengan 45 kali pengukuran.
English Abstract
The urea concentration can be determined by potentiometry using urea biosensor. Urea biosensor is made by coating the glass electrode with immobilized urease. The mass of immobilized urease affects the performance of urea biosensor. The performance of urea biosensor is showed by biosensor characteristics such as concentration range, detection limit, Nernstian factor, response time and biosensor efficiency. In this research, the optimation of immobilized urease mass in the range of 0.25g up to 2.00g was studied towards urea biosensor character. Biosensor character can be studied by measuring cell potential of urea solution in the range of 10 -6 M up to 1M. The results showed that the best perfomance of urea biosensor was gained at 1.00g immobilized urease mass, Nernstian factor 27.500 mV/decade, while the range of urea concentration can be measured from 10 -3 M up to 10 -1 M, the detection limit 0.38 10 -3 M, and biosensor response time 30 seconds. In addition the efficiency of urea biosensor can be used up to 45 time measurements.
