Abstract
Penelitian ini bertujuan untuk endeterminasi hubungan kekerabatan antara Nepenthes pectinata Danser dengan Nepenthes gymnamphora Nees. dan antar Nepenthes gymnamphora Nees. dari beberapa lokasi di pulau Jawa berdasarkan pola RFLP. Pengamatan pola RFLP dilakukan melalui tahapan isolasi DNA, pengukuran konsentrasi dan kemurnian dengan menggunakan NanoDrop spectrofotometer, pemotongan DNA dengan enzim EcoRI dan HaeIII dengan teknik RFLP serta elektroforesis gel agarosa untuk analisis DNA secara kualitatif. Hasil RFLP tersebut dianalisis dengan menggunakan program Clad 97 untuk mengetahui hubungan kekerabatan diantara Nepenthes. Pola fenogram hasil potongan EcoRI menunjukkan Nepenthes pectinata Danser berkerabat jauh dengan Nepenthes gymnamphora Nees. dan Nepenthes gymnamphora gunung Lemongan mempunyai jumlah pita paling banyak dibandingkan dua Nepenthes gymnamphora lainnya. Pola fenogram ini sama dengan fenogram hasil gabungan EcoRI dan HaeIII. Akan tetapi dari pola potongan HaeIII membentuk pola fenogram yang berbeda, hal ini diduga karena terjadi perbedaan area pada sekuen DNA yang dikenali oleh kedua enzim tersebut. Dari pola fenogram HaeIII terjadi indikasi kedekatan hubungan kekerabatan Nepenthes pectinata Danser dan Nepenthes gymnamphora Nees gunung Patuha.
English Abstract
The objective of this research was to determine the genetic relationship between Nepenthes pectinata Danser and Nepenthes gymnamphora Nees. and among Nepenthes gymnamphora Nees. from some locations in Java island based on RFLP pattern. The observation of RFLP pattern was done by the following steps: DNA isolation, DNA concentration and DNA purity measurement using NanoDrop Spectrophotometer, DNA digestion using EcoRI and HaeIII enzymes and visualization of DNA band using agarose gel electroforesis. The RFLP result was analyzed using Clad 97 program to determine the genetic relationship among Nepenthes. Phenogram pattern based on EcoRI digestion result showed that Nepenthes pectinata Danser has far relation with Nepenthes gymnamphora Nees., it also showed that Nepenthes gymnamphora from Lemongan mountain has the highest number of band compared to the other Nepenthes gymnamphora. This pattern was similar to the phenogram result based on the combination of digestion result of EcoRI and HaeIII. On the other hand, the pattern of HaeIII digestion result makes different pattern, it was estimated because of different site in DNA sequence which was recognized by both enzymes. The HaeIII pattern shows an indication of genetic relationship between Nepenthes pectinata Danser and Nepenthes gymnamphora Nees. from Patuha mountain.
