Abstract
Penelitian ini bertujuan untuk mempelajari pengaruh konsentrasi kitosan dalam amobilisasi selulase yang diisolasi dari Trichoderma viride. Aktivitas ekstrak kasar selulase amobil dapat dipelajari melalui penentuan kondisi optimum (pH, temperatur, waktu inkubasi) dan efisiensi pemakaian ulang. Ekstrak kasar selulase diamobilisasi dengan sistem double layer menggunakan Ca-alginat-kitosan. Aktivitas ekstrak kasar selulase amobil ditentukan melalui banyaknya glukosa hasil hidrolisis selulosa yang diukur secara spektrofotometri menggunakan metode Nelson-Somogyi. Hasil penelitian menunjukkan bahwa konsentrasi kitosan optimum untuk amobilisasi ekstrak kasar selulase adalah 2%. Kondisi optimum ekstrak kasar selulase amobil dicapai pada pH 5,5, temperatur 60 oC dan waktu inkubasi 20 menit, menghasilkan aktivitas sebesar 0,168 unit. Ekstrak kasar selulase amobil pada kondisi optimum masih layak dipakai sampai 4 kali pemakaian dengan efisiensi 58,24%.
English Abstract
The aim of this research is to study the influence of chitosan concentrations on cellulase immobilization which isolated from Trichoderma viride . Immobilized crude cellulase activity can be studied by determination of the optimum condition (pH, temperature, incubation time) and the efficiency of reuses of immobilized crude cellulase. Crude cellulase was immobilized in double layer system using Ca-alginate-chitosan. The activity of crude cellulase was determined through the number of glucose resulting from cellulose hydrolisis that was measured spectrofotometrically by using Nelson-Somogyi method. The result showed that the optimum chitosan concentration for crude cellulase immobilization was 2 %. The optimum condition of immobilized crude cellulase activity occurred at pH 5.5, temperature 60 oC and incubation time 20 minutes, yielding in 0.168 unit of activity. The immobilized crude cellulase at optimum condition can be used until four times within efficiency activity of crude cellulase was 58.24%.
