Abstract
Hiperglikemia dapat menyebabkan disfungsi sistem imun namun pengobatan hiperglikemia dengan obat sintetik dapat memberikan efek samping. Daun kersen (M. calabura L.) memiliki khasiat mengobati berbagai penyakit seperti hiperglikemia dan inflamasi. Tujuan dari penelitian ini yaitu mengetahui peran ekstrak daun kersen (EDK) terhadap produksi IL-6 oleh sel makrofag dan aktivitas NF-κB pada sel limfosit B dan sel limfosit NK pada mencit jantan BALB/c model hiperglikemia. Induksi hiperglikemia dilakukan menggunakan pakan lemak tinggi dan cairan fruktosa 20 %. EDK hasil dekoksi kering (pelarut akuades) diberikan pada mencit hiperglikemia secara oral dengan dosis 420, 700, dan 2800 mg/kg BB mencit selama dua minggu. Sel-sel imunokompeten diisolasi dari organ limpa kemudian dilakukan pewarnaan sel menggunakan antibodi dan analisis flow cytometry. Analisis data dilakukan dengan uji One-way ANOVA dan Post Hoc Tukey HSD pada signifikansi 95 % dengan program SPSS versi 16.0 for Windows. Hasil menunjukkan bahwa produksi IL-6 dan aktivitas NF-κB mengalami peningkatan pada mencit hiperglikemia. EDK pada ketiga dosis mampu menurunkan jumlah relatif sel makrofag yang memproduksi IL-6 dan sel limfosit B serta sel NK yang mengekspresikan NF-κB pada mencit hiperglikemia hingga kondisi normal. EDK mengandung flavonoid dan turunannya yang dapat mengurangi aktivitas NF-κB dan mengurangi produksi IL-6 melalui penghambatan aktivitas NF-κB. Secara keseluruhan, hasil menunjukkan bahwa EDK memiliki peranan penting dalam menghambat produksi IL-6 dan aktivitas NF-κB pada mencit BALB/c model hiperglikemia.
English Abstract
Hyperglycemia can cause immune system dysfunction but its treatment by synthetic drugs can cause the side effects for health. Cherry leaves (M. calabura L.) have the benefit to cure some diseases such as hyperglycemia and inflammation. This study aims to understand the role of cherry leaves extract (CLE) for IL-6 production by macrophages and NF-κB activity by B lymphocytes and Natural Killer (NK) cells in male BALB/c mice hyperglycemia model. Hyperglycemia induction was done by using high-fat diet and 20 % fructose. CLE made by dried decoction method (aqueous solvent) are used for oral treatment by doses 420, 700, and 2800 mg/kg weight body for two weeks. Immunocompetence cells are isolated from spleen and the cells are stained by specific antibody for flow cytometry analysis. Data analysis was done by using One-way ANOVA and Post Hoc Tukey HSD by 95 % significance on SPSS 16.0 for Windows program. The result shows that IL-6 production and NF-κB activity increased in hyperglycemic mice. CLE for three doses on hyperglycemia mice can decrease the relative number of macrophages that express IL-6 and B lymphocytes also NK cells that express NF-κB toward normal or health condition. CLE consists of some compounds such as flavonoid and its derivates that can decrease the NF-κB activity and IL-6 production by inhibiting the NF-κB activity. Overall, this study shows that CLE has an important role for decreasing IL-6 production and NF-κB activity on BALB/c mice hyperglycemia model.
