Abstract

Eceng Gondok (Eichhornia Crassipes) merupakan salah satu jenis tanaman yang tumbuh di air dengan tingkat pertumbuhan yang sangat cepat. Tanaman eceng gondok memiliki kandungan selulosa yang cukup besar yang dapat dimanfaatkan dalam hidrolisis enzimatik sehingga dihasilkan gula reduksi yang bisa digunakan dalam pembuatan bioetanol. Dalam pemanfaatan selulosa ini perlu dilakukan perlakuan awal (pretreatment) yang berguna untuk mengurangi kandungan lignin yang dapat menghambat proses hidrolisis dari selulosa. Proses pretreatment dengan iradiasi gelombang mikro mampu mendegradasi kandungan lignin dan hemiselulosa yang ada pada biomassa sehingga mampu menghasilkan gula reduksi yang lebih tinggi pada proses hidrolisis. Pada penelitian ini dilakukan pengamatan terhadap pengaruh perlakuan awal iradiasi gelombang mikro pada selulosa sebagai bahan (substrat) proses hidrolisis enzimatik. Enzim selulase kasar dari Trichoderma ressei dan Aspergillus niger digunakan sebagai katalis proses hidrolisis. Penelitian ini menggunakan metode Rancangan Acak Kelompok Faktorial (RAK) pola dua arah yang terdiri dari dua faktor yaitu faktor perlakuan pretreatment microwave dengan taraf (dengan pretreatment ; tanpa pre-treatment) dan faktor II waktu proses hidrolisis enzimatik dengan 4 taraf (4 jam, 8 jam, 12 jam, dan 16 jam). Dilanjutkan dengan analisis data dengan ANOVA digunakan selang kepercayaan BNT 1%. Proses pre-treatment microwave mampu menurunkan kadar lignin pada penelitian ini adalah sebesar 5,115 % (b/b). Enzim selulase yang dihasilkan dari mikroorganisme viii Trichoderma ressei dan Aspergillus niger memiliki nilai aktivitas enzim sebesar 0,96325 IU/ml. Gula reduksi tertinggi yang dihasilkan adalah sebesar 0,088 % (b/v) yaitu pada perlakuan dengan pre-treatment microwave dan pada waktu hidrolisis 4 jam. Faktor dengan perlakuan pre-treatment microwave sangat berpengaruh nyata terhadap hasil gula reduksi pada proses hidrolisis. Faktor perlakuan waktu hidrolisis sangat berpengaruh nyata terhadap hasil gula reduksi pada proses hidrolisis.

English Abstract

Water hyacinth (Eichhornia crassipes) is a kind of aquatic plants that have high growth rates. Water hyacinth has huge cellulose content which could be utilized in enzymatic hydrolysis process to produce reducing sugars that can be used in advanced process such as bioethanol fermentation. In the utilization of cellulose, pretreatment process is necessary to reduce lignin content in the material that can inhibit the enzymatic hydrolysis process of cellulose. The pretreatment process with microwave irradiation can degrade lignin and hemicellulose contained in the biomass so as be able to produce higher number of reducing sugars in enzymatic hydrolysis process. This study is carried out observation about the effect of microwave irradiation pretreatment of cellulose as a material (substrate) in enzymatic process. Crude Cellulase enzymes produced by Trichoderma reesei and Aspergillus niger is used as catalyst in hydrolysis process. This study uses Factorial randomized block Design in two directions pattern which consists of two factors. The first factor is pretreatment process in material (not pretreated material and pretreated material). The second factor is the time of hydrolysis process which consists of four levels (4 hours, 8 hours, 12 hours, and 16 hours). It is used ANOVA in analyze with BNT significance level 1 % so as the effect of each factors can be known. In this study, microwave pretreatment process can degrade lignin content in material by 5,115% (w/w). The Enzyme activities level of Cellulase enzymes that is produced by microorganisms Trichoderma reesei and Aspergillus niger was calculated by 0,96325 IU/ml. The highest reducing sugar content x produced is amounted to 0,088% (w/v) that is on the microwave pretreated material and 4 hours enzymatic hydrolysis time process. Both factors those are the microwave pretreatment and the hydrolysis time process have a very significant effect in the result of reducing sugar in hydrolysis process.
