Abstract
Hiperkolesterolemia merupakan faktor risiko penyakit jantung koroner. Pencegahan peningkatan kolesterol darah dapat dilakukan dengan mengkonsumsi pangan fungsional berbahan dasar herbal. Tanaman cincau hitam (Mesona palustris Bl) dipercaya mampu menjadi solusi perawatan, penurunan, penyembuhan, bahkan sebagai proteksi organ dalam dari efek negatif hiperkolesterolemia. Banyak peneliti yang telah mengembangkan untuk manfaat kesehatan dari produk berbasis cincau hitam . Penelitian ini bertujuan untuk mengetahui efektivitas gel cincau hitam di pasaran terhadap pencegahan peningkatan kolesterol darah serta karakteristik fisik dan kimianya. Penelitian menggunakan 2 tahap. Tahap 1 adalah sampling gel cincau hitam pasaran dari 3 tempat yang berbeda yaitu pasar tradisional di Lawang dan di Malang, and supermarket di Malang. Selanjutnya ketiga cincau tersebut akan dianalisa kadar antioksidan, total fenol, total padatan terlarut (TPT), tekstur, dan warna. Berdasakan metode Zeleny, gel cincau hitam terbaik adalah gel cincau hitam dari supermarket di Malang. Gel cincau hitam dari supermarket mempunyai karakteristik kadar antioksidan sebesar 68,51%, total fenol 197 ppm, kekuatan tekstur sebesar 25,7 N/m2, TPT sebesar 3,63%, hasil analisa warna L*= 26,47, a= 1,63, b= 1,1, dan analisa antioksidan IC50 sebesar 133,964 ppm. Berdasarkan hasil di tahap ini, gel cincau hitam dari supermarket digunakan untuk pencegahan peningkatan kolesterol darah pada tikus. Desain penelitian yang digunakan pada penelitian tahap 2 adalah pre post only control group design dengan nested design.Tikus akan dibagi menjadi 5 kelompok perlakuan dengan 2 kontrol dan 3 perlakuan. Kelompok 1 yaitu kontrol negatif hanya diberi pakan standart. Kelompok 2 adalah kontrol positif yang diberikan pakan tinggi kolesterol. Kelompok 3,4, dan 5 diberikan pakan tinggi kolesterol dan gel cincau hitam dengan dosis secara berurutan 1,8g/ 200g BBT. 3,6g/200g BBT, dan 5,4g/200g BBT. Data hasil pengamatan akan diolah secara statistik menggunakan ANOVA dengan selang kepercayaan 1%. Hasil penelitian menunjukkan bahwa perlakuan pemberian gel cincau hitam memberikan pengaruh nyata (α=0.01) terhadap pencegahan peningkatan kolesterol. Kelompok 5 menunjukkan pencegahan peningkatan paling tinggi dari minggu ke-0 hingga minggu ke-3. Pada dosis ini mampu menurunkan kadar kolesterol sebesar 12,79%, kadar trigliserida sebesar 58,755%, dan kadar LDL menurun sebesar 61,970% namun kadar HDL meningkat sebesar 22,322%,
English Abstract
Hypercholesterolemia is a risk factor for coronary heart disease. Prevention of an increase in blood cholesterol can be done by consuming herbal-based functional foods. Black grass jelly (Mesona palustris Bl) is believed to be decrease, healing, even as an organ protection from negative effect of hypercholesterolemia. Many researchers have developed for health benefits black grass jelly based products health benefits. This study aims to determine the effectiveness of commercial black grass jelly gel to prevent an increasing blood cholesterol and knowledge of physic and chemical characteristic. This study used two stages. First stage was sampling of black grass jelly gel from 3 different, traditional markets in Lawang and in Malang and supermarket in Malang. The black grass jelly were analyzed the antioxidant content, total phenol, total dissolved solids (TPT), texture, and color. Basic on the Zeleny method, the best of black grass jelly gel obtained from supermarket in the Malang. This black grass jelly gel has characteristic of a 68,51% of antioxidant levels, 197 ppm of phenol, 25,7 N/m2 of texture force, 3,63% of TPT, color analysis L*= 26,47, a= 1,63, b= 1,1, and the analysis of antioxidant IC50 of 133,964 ppm. Furthermore this stage, black grass jelly gel was used to prevent an increasing blood cholesterol in rats. The research design used a pre post-only control group design with nested design.The rats were divided into 5 groups consisting 2 controls and 3 treatments. Group 1 of negative control was treated with only standart feed. Group 2 of positive control was treated with high cholesterol feed. Group 3,4, and 5 were treated with high cholesterol feed and black grass jelly gel dose of 1,8g/200g, 3,6g/200g, and 5,4g/200g body weight, respectively. Data was processed using ANOVA with a confidence interval of 1%. The results showed that treatments of black grass jelly gel gave significant effect (α = 0.01) in the prevention of an increasing in cholesterol. Group 5 gave the highest decreasing of blood cholesterol observed from week-0 till week-3. This dose decreased cholesterol by 12,79%, decreased triglyceride levels by 58,755%, and LDL levels decreased by 61,970% but increased HDL levels by 22,322%
