Abstract
Konsumsi minyak goreng nasional sebesar 3,2 juta ton dan sebanyak 63% adalah minyak curah. Dengan meningkatnya produksi dan konsumsi minyak goreng, ketersediaan minyak jelantah semakin melimpah. Tingginya radikal bebas pada minyak jelantah akan menyebabkan oksidasi sehingga memicu kerusakan jaringan dan sel pada organ hati, sehingga enzim transaminase yaitu enzim SGPT (Serum Glutamat Piruvat Transaminase) dan SGOT (Serum Glutamat Oksaloasetat Transaminase) yang ada pada organ hati akan keluar dan masuk ke peredaran darah, sehingga dengan pemeriksaan biokimia pada serum akan timbul kenaikan SGOT dan SGPT yang tinggi. Antioksidan merupakan zat yang dapat menetralkan radikal bebas dengan cara menyumbangkan satu atom hidrogen dari gugus OH kepada lipid peroksida yang bersifat radikal sehingga menjadi radikal bebas yang kurang reaktif dan tidak merusak sehingga enzim SGPT dan SGOT tidak keluar dalam peredaran darah. Salah satu sumber antioksidan alami yaitu kulit buah naga merah dan rosella. Berdasarkan latar belakang di atas, diperlukan suatu produk fungsional yang dapat berfungsi sebagai antioksidan alami sehingga dapat menurunkan kadar SGPT dan SGOT dalam darah. Jelly drink kulit buah naga merah dan rosella dipilih sebagai minuman fungsional yang tidak hanya berfungsi sebagai penunda lapar, tapi juga sebagai antioksidan alami Formulasi terbaik dari Jelly Drink Kulit Buah Naga Merah dan Rosella serta penambahan bahan pengental dan gula stevia yang merupakan gula rendah kalori selanjutnya akan diuji secara in vivo untuk mengetahui pengaruhnya terhadap kadar SGOT dan SGPT tikus wistar jantan yang diinduksi minyak jelantah. Penelitian ini dilakukan melalui 2 tahap. Tahap I menentukan formulasi terbaik jelly drink kulit buah naga merah dan rosella menggunakan Rancangan Acak Kelompok (RAK) terdiri dari 2 faktor, Faktor I = Formulasi Sari Kulit Buah Naga dan Rosella dan faktor II = penambahan karagenan : glukomanan. Masing- masing diulang sebanyak 3 kali sehingga didapat 18 satuan percobaan. Tahapan II yaitu pengujian secara in vivo pengaruh jelly drink kulit buah naga dan rosella terhadap kadar kadar SGOT dan SGPT dengan desain penelitian Pre and Post Test with Control Group dan pemilihan objek penelitian untuk pengelompokan dan pemberian perlakuan menggunakan rancangan tersarang (Nested design) dengan 2 faktor. Pemilihan perlakuan terbaik menggunakan metode Zeleny. Analisa keragaman diuji dengan ANOVA, beda nyata antar perlakuan dilanjutkan dengan uji DMRT atau BNT 5%. Hasil penelitian yang telah dilakukan jelly drink kulit buah naga dan rosella dengan nilai IC50 sebesar 196,26, total antosianin sebesar 68,076 dari penentuan perlakuan terbaik Metode Zeleny juga memberikan nilai yang paling rendah sebesar 0,035. Selain itu, penelitian secara in vivo menunjukkan bahwa perlakuan jelly drink kulit buah naga dan rosella memberikan pengaruh nyata (α=0,05) terhadap penurunan kadar SGOT dan SGPT tikus wistar jantan. Hasil SGOT dan SGPT paling besar diberikan oleh pengaruh perlakuan pemberian jelly drink dosis 2 (7,2ml/200g BB). Penurunan kadar SGOT selama 2 minggu pemberian jelly drink sebesar 24,99% dan kadar SGPT 32,74 %.
English Abstract
National oil consumption by 3.2 million tons and as much as 63% are bulk oil. With the increased production and consumption of cooking oil, waste cooking oil availability of more abundant Free radical are high on used oil and it will cause damage on tissues and cells in the liver, so transaminase enzyme, that is SGPT enzyme (Serum Glutamic Pyruvate Transaminase) and SGOT enzyme (Serum Glutamate Oxaloacetate Transaminase) that exist in the liverwill come out and enter the blood circulation, and with biochemical examination there will be high increase SGOT and SGPT in serum. Antioxidant is substances that can neutralize free radical by donating one hydrogen atom from OH group to the peroxide lipid which is radical, so free radical becomes less reactive and not destructive, and it makes SGPT and SGOT enzyme do not come out from the blood circulation. One source of natural antioxidant is peel of red dragon fruit and rosella. Based on above explanation, we need functional product which can used as natural antixodant so it can help to reducing levels of SGPT and SGOT in the blood. Jelly drink from peel of red dragon fruit and rosella has chosen as a functional beverage that not only served as hungry procrastinators, but also as a natural antioxidant. Best formulation from jelly drink and the addition of thickening agents and stevia sugar which is a low calorie sugar then will tested in in vivo for determine the effect of jelly drink to levels of SGOT and SGPT in male wistar rats that induced by used cooking oil. This research was conducted through two stages. First stage is determine the best formulation jelly drink from skin of red dragon fruit and rosella using a Randomized Block Design (RBD) consisting of two factor, Factor I = Formulation of filtrate skin of red dragon fruit and rosella, Factor II = addition of carrageenan : glucomannan. Each formula repeated 3 times so that got 18 experimental units. Second stage is test in in vivo to know the effect of jelly drink from peel of red dragon fruit and rosella against the level of SGOT and SGPT using Pre and Post Test with Control Group experiment design and object selection for grouping and giving treatment is using Nested Design with 2 factors. Selection of the best treatment is using Zeleny method. Analysis of diversity were tested by ANOVA, if there is a significant difference in the treatments then continued with DMRT or BNT 5%. Results of this research in jelly drink from peel of red dragon fruit and rosella have IC50 value with 196,26 ppm, total anthocyanin 68,076 ppm from the best treatment in zeleny method also gives the lowest score of 0.035. In addition, in vivo research show that treatment with jelly drink from skin of red dragon fruit and rosella have significant effect (α=0.05) to decrease the level pf SGOT and SGPT in male Wistar rats. Decreased SGOT and SGPT levels is given by treatment of 2 doses jelly drink (7.2 ml/200 g BB). Decreased level of SGOT and SGPT in 2 weeks giving jelly drink was 24.99% and 32.74%.
