Abstract
E. coli menghasilkan endotoksin yang menyebabkan gastroenteritis. Gastroenteritis ditandai dengan adanya inflamasi pada membran mukosa saluran pencernaan dan ditandai dengan diare dan muntah. Daun sawo (Manilkara zapota L.) mengandung flavonoid dan tannin yang berfungsi sebagai antibakteri dan antioksidan. Penelitian ini bertujuan untuk mengetahui efek terapi ekstrak daun sawo manila (Manilkara zapotta) terhadap gastroenteritis pada tikus putih (Rattus norvegicus) induksi E. coli berdasarkan kadar Malondyaldehide (MDA) dan histopatologi kolon. Rancangan penelitian ini bersifat eksperimental dengan post test control only menggunakan rancangan acak lengkap (RAL). Penelitian menggunakan hewan model tikus putih (Rattus norvegicus) jantan berumur 5-6 minggu yang dibagi menjadi 5 kelompok, yaitu: kelompok kontrol negatif, kelompok kontrol positif. Kelompok terapi ekstrak etanol daun sawo dengan dosis 200 mg/kg BB, 300 mg/kg BB dan dosis 400 mg/kg BB. Kadar MDA diukur dengan metode TBA dan histopatologi kolon menggunakan pewarnaan HE (Hematoksilin-Eosin). MDA dianalisis dengan one way ANOVA dengan tingkat kepercayaan = 0.05 dan histopatologi kolon dianalisis secara deskriptif. Hasil penelitian menunjukkan pemberian terapi ekstrak etanol daun sawo dosis 400 mg/kg BB merupakan dosis terbaik yang mampu menurunkan kadar MDA organ kolon serta mampu memperbaiki kerusakan mukosa kolon ditandai dengan tidak adanya erosi epitel serta menurunnya infiltrasi sel radang. Disimpulkan bahwa ekstrak etanol daun sawo manila dapat digunakan sebagai terapi gastroenteritis.
English Abstract
E. coli produce endotoxin which cause gastroenteritis. Gastroenteritis is inflammation of the mucous membrane of the digestive tract and is characterized by diarrhea and vomit. The leaves of sawo tree (Manilkara zapota L.) are contain chemical compounds, flavonoids and tannins that act as antibacteria and antioxidant. This research aims to know therapeutic effects of extracts from manilkara leaves (Manilkara zapota L.) against gastroenteritis in male white rats (Rattus norvegicus) induced Escherichia coli as seen from Malondialdehyde (MDA) level and colon histopathology. This research used RAL experimental with post test control design only. This research using animal model, the male white rat (Rattus norvegicus) was 5-6 weeks are divided into 5 groups: were negative control group, gastroenteritis group, and three groups gastroenteritis rats with terapy ethanol extracts of Manilkara leaves are given for 8 days dose 200mg/kg BW, 300 mg/kg BW, and 400 mg/kg BW. MDA levels were measured by TBA method and colon histopathology using HE staining (Hematoxylin-Eosin). MDA level analyzed by one way ANOVA with a confidence level is a α = 0.05 and colon histopathology analyzed are descriptively. The result showed that the ethanol extracts therapy dose of 400 mg/kg BW is the best dose because that dose could decreas MDA levels the colon and that dose could repair damage on colon mucosal and decreas the inflammatory cell in gastroenteritis rats. The conclusion of this study is ethanol extracts of manila leaves could use as gastroenteritis therapy.
