Abstract

Rhodamin B merupakan zat toksik dan sumber radikal bebas apabila masuk kedalam tubuh. Zat aktif Rhodamin dapat berikatan dengan lemak tidak jenuh pada membrane sel hepar dan menyebabkan terjadinya peroksidase lipid yang berujung pada kerusakan sel hepar. Hasil akhir dari peroksidase lipid ini adalah Malondialdehyde. Untuk menghambat terjadinya kerusakan sel akibat radikal bebas ini diperlukan antioksidan seperti isoflavon yang ada dalam bawang putih. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian perasan air bawang putih terhadap kerusakan sel hepar melalui gambaran histopatologi dan pengukuran kadar MDA pada tikus (Rattusnorvegicus) yang dipapar rhodamin melalui pakan selama 21 hari. Penelitian ini terdiridari lima kelompok perlakuan yaitu kelompok control negatif (A), kelompok control positif (B), kelompok tikus C, D dan E yang diberikan pakan mengandung Rhodamin B dan diberikan perasan air bawang putih sebanyak 0,5 ml/200gBB, 1 ml/200gBB, dan 1,5 ml/200gBB secara bersamaan. Pengukuran kadar MDA hepar dilakukan menggunakan spektofotometer UV VIS sedangkan pengamatan histopatologi hepa rmenggunakan pewarnaan Hematoxylen-Eosin (HE). Analisa data MDA menggunakan One Way ANOVA dan uji lanjutan BNJ α 5% serta histopatologi hepar secara deskriptif. Hasil penelitian menunjukkan air perasan bawang putih memberikan pengaruh nyata (p<0,01) terhadap kadar MDA histopatologi hepar. Air perasan bawang putih dengan dosis terapi 0,5 ml/200gBB merupakan volume palig efektif dalam menurunkan kadar MDA pada hepar dengan prosentase 40% dan mencegah kerusakan gambaran histopatologi hepar.

English Abstract

Rhodamin B is a toxic substances and source of the free radicals if enter the body. Rhodamin B in feed that enter the body will be absorbed in the intestinal villi and circulated with blood to the liver for detoxification. Rhodamin B that is a free radicals substances binding with the unsaturated fats in the liver membrane cell and going on lipid peroxidation which lead to liver damage. The end result of lipid peroxidation is Malondialdehyde (MDA). To prevent the occurrence of liver cell damage, can consume food that contain antioxidant. One of the antioxidant compound is isoflavones that can be found in garlic juice . This study was aimed to evaluate the MDA level and histopathology of the liver tissue in rats with rhodamine exposure in feed after giving different volumeswater of juice of garlic. In this study, these animals were devided into five groups, group A was negative control, group B was positive control, and grup C, D, and E treated with borax in feed and received water juice of garlic will different volume (0,5 ml/200gBW 1 ml/200gBW, and 1,5 ml/200gBW) simultaneously. Measurement of MDA level performed using UVspectrophotometer. And the histopathology of liver was observedusingHematoxylen-Eosin (HE) staining. The MDA data will be analisis using One Way ANOVA and BNJ test α 5%, histopathology of liver and hepar histopathology will be analysed descriptivity.The results showed that garlic juice had a real effect (p <0.01) on MDA level of histopathologic hepar. Garlic juice with a dose of 0.5 ml / 200gBB therapy is an effective volume in reducing MDA levels in the liver by 40% and preventing damage to hepatic histopathologic features.
