Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit peradangan pada organ saluran pencernaan termasuk lambung. Salah satu faktor penyebab IBD disebabkan oleh induksi obat-obatan NSAID seperti indometasin. Hal ini disebabkan mekanisme kerja indometasin menghambat kinerja enzim COX (siklooksigenase) yang mengakibatkan kenaikan ROS pada jaringan lambung dan menimbulkan produksi sitokin proinflamatori seperti TNF-α. Daun ubi jalar ungu (Ipomoea batatas L.Lam) kaya akan kandungan flavonoid dan antosianin yang berperan sebagai antioksidan dan antiinflamasi. Kandungan tersebut berpotensi dalam menekan ekspresi TNF-α serta memperbaiki kerusakan jaringan organ lambung akibat penyakit IBD. Penelitian ini dilakukan dengan memberikan terapi ekstrak etanol daun ubi jalar ungu (dosis: 600 mg/kgBB, 700 mg/kgBB dan 800 mg/kgBB) pada tikus (Rattus norvegicus) model IBD yang diinduksi dengan indometasin ( dosis: 15 mg/kgBB). Parameter yang diamati pada penelitian ini berupa ekspresi TNF-α menggunakan metode imunohistokimia dan histopatologi organ lambung yang diwarnai dengan pewarna HE (Hematoxyline-eosin). Sedangkan analisia data hasil penelitian dilakukan menggunakan metode ANOVA (Analysis of Variance) dan dilanjutkan dengan uji lanjutan BNJ (Beda Nyata Jujur) α-0,05. Hasil penelitian ini menunjukan bahwa pemberian ekstrak ethanol daun ubi jalar ungu pada tikus putih (Rattus norvegicus) model IBD induksi indometasin, dengan dosis 600 mg/kgBB, 700 mg/kgBB dan 800 mg/kgBB dapat mengurangi ekspresi TNF-α, perbaikan gambaran histopatologi lambung serta penurunan infiltrasi sel radang. Dosis terapi terbaik pada penelitian ini adalah pada dosis 800 mg/kgBB. Kesimpulan dari penelitian ini adalah ekstrak ethanol daun ubi jalar ungu dapat dijadikan terapi untuk IBD
English Abstract
Inflammatory Bowel Disease (IBD) is one of the gastrointestinal tract disorders including the stomach. One of the factors causing IBD is the induction of NSAID such as indomethacin. This is due to the indomethacin action that inhibiting COX enzymes (cyclooxygenase). That resulting in the increase of ROS level in tissues and initiate production and increase expression of proinflammatory cytokines such as TNF-α. This, resulting in neutrophil aggregation and mucosal damage to gastric organs. Purple sweet potato leaves (Ipomoea batatas L.Lam) are rich in flavonoids and anthocyanins that act as antioxidants and anti-inflammatories. It has potential to suppress TNF-α expression as well as repair damaged gastric tissue resulting from IBD. This study was performed by giving ethanol extract of purple sweet potato leaves (doses: 600mg/kgBB, 700mg/kgBW, and 800mg/kgBW) to Inflammatory bowel disease rats (Rattus norvegicus) induced by indomethacin (15mg/kg BW). The parameters observed in this study are TNF-α expression using immunohistochemical method and histopathology of stomach organ stained with HE (Hematoxyline-eosin). Data analysis of this research were conducted using ANOVA (Analysis of Variance) and continued with HSD test (Honestly Significant Difference) α-0,05. The data results showed that ethanol extract of purple sweet potato leaves in white rat (Rattus norvegicus) IBD indometacin induction model, with dose of 600 mg / kg BW , 700 mg / kg BW and 800 mg / kg BW could decrease expression of TNF-α and stomach histopathology improvement and decrease infiltration of inflammatory cells. However, the best therapeutic dose in this study was 800 mg / kgBW. The conclution is ethanol extract of purple sweet potato leaves can be used as therapy for IBD
