Abstract

Aflatoksin merupakan metabolit sekunder yang diproduksi oleh kapang yaitu Aspergillus flavus. Komponen aflatoksin dapat mengganggu fungsi filtrasi glomerulus ginjal yang dapat mempengaruhi parameter kimiawi ginjal yaitu BUN dan kreatinin. Bakteri asam laktat mempunyai kemampuan mengikat aflatoksin karena memiliki komponen asam teichoic pada dinding sel. Penelitian ini bertujuan untuk mengetahui efek bakteri Lactobacillus plantarum terhadap level BUN dan kreatinin serum serta histpatologi ginjal tikus (Rattus norvegicus) yang diinduksi aflatoksin. Penelitian ini dilakukan secara eksperimental menggunakan RAL. Hewan coba dibagi menjadi 5 kelompok yang terdiri dari kelompok kontrol negatif tanpa diberi perlakuan, kelompok aflatoksikosis yang diinduksi aflatoksin (7 µg/kg BB), kelompok P1 (BAL 1x104 CFU/ml dan aflatoksin 7 µg/kg BB), kelompok P2 (BAL 1x106 CFU/ml dan aflatoksin 7 µg/kg BB), dan kelompok P3 (BAL 1x108 CFU/ml dan aflatoksin 7 µg/kg BB). Level BUN dan kreatinin diukur menggunakan spektrofotometer secara kuantitatif dengan uji statistik One-Way ANOVA, α = 5% dilanjutkan dengan uji Beda Nyata Jujur (BNJ). Histopatologi ginjal menggunakan pewarnaan Hematoksilin-Eosin (HE) dan dianalisa secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian BAL dapat mencegah kenaikan BUN dan kreatinin serum serta mencegah kerusakan glomerulus dan tubulus ginjal akibat induksi aflatoksin secara signifikan (p<0,05). Dosis 1x104 CFU/ml adalah dosis efektif dalam mencegah kenaikan BUN dan kreatinin serum serta kerusakan glomerulus dan tubulus ginjal. Kesimpulan penelitian ini adalah pemberian BAL Lactobacillus plantarum dapat mencegah kenaikan level BUN dan kreatinin serum serta mencegah kerusakan ginjal.

English Abstract

Aflatoxin is a secondary metabolite produced by mold such as Aspergillus flavus. Components of Aflatoxin can interfere with the function of the kidney glomerulus filtration which can affect the chemical parameters of the kidney on BUN and creatinine. Lactic acid bacteria has the ability to bind aflatoxin because it has the teichoic acid component in the cell wall. The purpose of this study was to know the effect of Lactobacillus plantarum bacteria on BUN and creatinine serum levels and histopathology of the kidney in rats induced by aflatoxin. This research was conducted experimentally using RAL. The animals were divided into 5 groups consisted of negative control group without treatment, aflatoxicosis group induced aflatoxin (7 µg/kgBW), group P1 (BAL 1x104 CFU/ml and aflatoxin 7 µg/kg BW), group P2 (BAL 1x106 CFU/ml and aflatoxin 7 µg/kg BW), and group P3 (BAL 1x108 CFU/ml and aflatoxin 7 µg/kg BW). BUN and Creatinine levels were measured quantitatively using spectrophotometer and was analyzed statistically by One-Way ANOVA method, α = 5% followed by Bright Interpretation (BNJ). The histopathologic of the kidney used the HematoxylinEosin (HE) and was analyzed descriptively. The results showed that the administration of BAL could prevent an increase in BUN and serum creatinine and prevent damage to the glomerulus and kidney tubules due to induction of aflatoxin exposure significantly (p <0.05). A dose of 1x104 CFU/ml was an effective dose in preventing BUN and creatine serum and a few glomerulus and tubules damage of the kidney. The conclusion of this study was Lactobacillus plantarum could prevent increase of the BUN and creatine levels and kidney damage.
