Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit peradangan pada saluran pencernaan seperti duodenum, jejunum, ileum dan kolon yang menyerang hewan dan manusia. Salah satu faktor penyebab penyakit ini adalah induksi obatobatan NSAID seperti indometasin. Daun ubi jalar ungu mengandung senyawa flavonoid, polifenol dan antosianin. Penelitian ini bertujuan untuk mengetahui efek terapi ekstrak etanol daun ubi jalar ungu terhadap ekspresi TNF–α dan histopatologi ileum tikus model IBD. Model IBD diperoleh dari induksi indometasin 15 mg/kg/BB secara per oral. Penelitian menggunakan tikus putih (Rattus norvegicus) jantan berumur 8-12 minggu, dibagi menjadi 5 kelompok penelitian yaitu kontrol negatif, kontrol positif, dosis terapi 600, 700 dan 800 mg/kg/BB. Pemberian ekstrak etanol daun ubi jalar ungu diberikan selama 7 hari secara peroral. Pengamatan Ekspresi TNF–α menggunakan teknik imunohistokimia dan perubahan histopatologi ileum dengan pewarnaan HE. Perubahan histopatologi ileum berupa data kualitatif yang dianalisa secara deskriptif. Selain itu, ekspresi TNF–α dianalisa secara kuantitatif dengan menggunakan program Immuno Ratio dan dilanjutkan dengan metode One Way Analysis of Variance (ANOVA) dilanjutkan dengan uji Tukey dengan α = 0,05. Hasil menunjukkan bahwa pemberian ekstrak etanol daun ubi jalar ungu pada tikus model IBD mampu menurunkan ekspresi TNF–α paling baik sebesar 74,73% dan perbaikan sel epitel, sel goblet serta penurunan infiltrasi sel radang.
English Abstract
Inflammatory Bowel Disease is digestive disorder caused inflammation in duodenum, jejunum, ileum and colon that attacks animal and humans. Inflammatory Bowel Disease also caused by NSAID drugs such as indomethacin. Purple sweet potato leaves contain flavonoid, polyphenols and anthocyanin compounds. This study aims to determine the effect of purple sweet potato leaves extract on TNF-α expression and histopathology ileum on rats IBD model. Rats of IBD model were obtained from indomethacin induction as much as 15 mg / kg / BW orally. The research used male rats (Rattus norvegicus) aged 8-12 weeks and divided into 5 groups, they are negative control group, positive control group, therapy group with doses 600, 700 and 800 mg / kg / BW. Ethanol extract of purple sweet potatoes leaves given for 7 days orally. TNF-α expression determined by immunohistochemistry and ileum histopathology stained by HE stain. Histopathology of ileum was analyzed descriptively. In addition, TNF-α expression was analyzed quantitatively with Immuno Ratio programing continued by One Way Analysis of Variance (ANOVA) and continued with Tukey test with α = 0.05. The results showed that ethanol extract of purple sweet potato leaves was able to decrease TNF-α expression at best 74.73% and repair of epithelial cell, goblet cell and decreased infiltration of inflammatory cells.
