Pengaruh Ekstrak Daun Andong (Cordyline fruticosa) dalam Menghambat Pertumbuhan Bakteri Aggregatibacter actinomycetemcomitans secara In Vitro

Abstract

Penyakit periodontal merupakan penyakit dalam rongga mulut yang diderita oleh hampir semua manusia di dunia dan mencapai 50% dari jumlah populasi orang dewasa. Salah satu faktor penting yang menyebabkan penyakit periodontal adalah bakteri patogen Aggregatibacter actinomycetemcomitans. Andong (Cordyline fruticosa) adalah tanaman hias yang jumlahnya sangat melimpah di Indonesia dan sejak dulu digunakan sebagai ramuan herbal dalam menghentikan perdarahan. Daun Andong (Cordyline fruticosa) mengandung banyak senyawa aktif diantaranya saponin, tanin, flavonoid, polifenol, steroida, polisakarida, kalsium oksalat, dan zat besi. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak daun andong (Cordyline fruticosa) dalam menghambat pertumbuhan bakteri Aggregatibacter actinomycetemcomitans secara in vitro. Penelitian ini dilakukan dengan rancangan true experimental design post test control only. Terdapat 7 kelompok perlakuan yang terdiri ekstrak daun andong (Cordyline fruticosa) dengan konsentrasi 6,25%, 12,5%, 25%, 50%, 100%, kontrol negatif dengan menggunakan akuades steril, dan kontrol positif dengan menggunakan klorheksidin glukonat 0,2%. Berdasarkan uji statistik one way Anova (p<0,05), didapatkan perbedaan bermakna antara zona hambat bakteri Aggregatibacter actinomycetemcomitans pada setiap kelompok perlakuan. Selanjutnya, dari uji korelasi Pearson didapatkan adanya hubungan yang sangat erat dan berbanding lurus antara besarnya konsentrasi dengan diameter zona hambat bakteri Aggregatibacter actinomycetemcomitans. Dari penelitian ini dapat disimpulkan adanya pengaruh ekstrak daun Andong (Cordyline fruticosa) dalam menghambat pertumbuhan bakteri Aggregatibacter actinomycetemcomitans secara in vitro.

English Abstract

Periodontal disease is a disease in the oral cavity suffered by almost all humans in the world and reaches 50% of the adult population. One important factor that causes periodontal disease is the bacterial pathogen Aggregatibacter actinomycetemcomitans. Andong (Cordyline fruticosa) is an abundant ornamental plant in Indonesia and has been used as an herb in stopping bleeding. The Andong (Cordyline fruticosa) leaves contain many active compounds such as saponins, tannins, flavonoids, polyphenols, steroids, polysaccharides, calcium oxalate, and iron. The purpose of this study was to determine the effect of andong (Cordyline fruticosa) leaves extract in inhibiting growth of Aggregatibacter actinomycetemcomitans bacteria in vitro. The design of the research used is true experimental design post test control only. There are 7 groups consists of andong (Cordyline fruticosa) leaves extract within concetration 6.25%, 12.5%, 25%, 50%, 100%, negative control group with sterile aquadest, and positive control group with Chlorhexidine gluconate 0.2%. Based on the statistical test of one way Anova (p <0.05), there was significant difference between the obstacles zone diameter of Aggregatibacter actinomycetemcomitans bacteria in each control group. Furthermore, from Pearson correlation test found a very close relationship and directly propotional between the amount of concentration with the obstacles zone diameter of Aggregatibacter actinomycetemcomitans bacteria. From this research concluded that there is effect of andong (Cordyline fruticosa) leaves extract in inhibiting growth of Aggregatibacter actinomycetemcomitans bacteria in vitro.
