Abstract
Penggunaan obat nyamuk berbahaya karena memiliki kandungan bahan aktif d-allethrin menyebabkan gangguan pada pernafasan. Obat nyamuk mengandung 10 molekul radikal dalam satu kali hisapan yang dapat memicu kerusakan pada sel alveolar. Buah semangka kandungan antioksidannya berupa likopen pada bagian buah yang tinggi jika dibandingkan buah dan sayuran lainnya. Sehingga buah semangka dapat dijadikan alternatif sebagai tindakan preventif dalam menangkal radikal bebas pada tikus (Rattus norvergicus) yang di papar obat nyamuk. Penelitian ini bertujuan untuk mengetahui efek preventif ekstrak buah semangka terhadap ekspresi IL-6 dengan gambaran histopat alveolar. Penelitian ini menggunakan metode Rancangan Acak Lengkap, terdiri dari lima kelompok yaitu kelompok kontrol negatif, kelompok kontrol positif yang dipapar menggunakan obat nyamuk bakar selama 4 jam/hari selama 20 hari, kelompok perlakuan 1, perlakuan 2 dan perlakuan 3 yaitu tikus putih yang dipapar asap obat nyamuk, sebelumnya pemaparan diberikan ekstrak semangka masing-masing dengan dosis sebesar 25 mg/kg BB, 50 mg/kg BB, dan 100 mg/kg BB selama 20 hari. Parameter yang diamati adalah ekspresi IL-6 menggunakan metode Imunohistokimia dan histopatologi alveolar menggunakan pewarnaan Hematoxylin Eosin. Analisa data ekspresi IL-6 dilakukan secara statistik kuantitatif dengan metode ANOVA dilanjutkan dengan BNJ (α=0,05), sedangkan gambaran histopatologi alveolar dianalisa secara deskripftif. Hasil penelitian menunjukkan bahwa pemberian preventif buah ekstrak semangka secara signifikan mampu menurunkan ekspresi IL-6 dan dapat mencegah kerusakan gambaran histopatologi alveolus. Dosis 100 mg/kgBB merupakan dosis terbaik yang mampu menunjukkan hasil mendekati kelompok Kontrol Negatif. Kesimpulan dari penelitian ini adalah Buah semangka (Citrullus lanatus) dapat digunakan sebagai preventif pada tikus yang diberikan paparan asap obat nyamuk.
English Abstract
Use mosquito repellent because it has d-allethrin active ingredients causing respiratory disturbance. The mosquito repellent contains 10 radical molecules in one suction that can point to damage to alveolar cells. Watermelon fruit contains antioxidant substances in the fruit is high when compared to other fruits and vegetables. Because watermelon fruit can be used as an alternative to preventive measures in warding free radicals in mice (Rattus norvergicus) in the mosquito repellent. This study aims to determine the effect of watermelon extract prevention on IL-6 expression by modifying alveolar histopaths. This study used the Completely Randomized Design Method, consisting of five groups: negative control group, positive control group exposed using mosquito coils for 4 hours / day for 20 days, treatment group 1, treatment 2 and treatment 3 white mice were immediately exposed to mosquito coils, previously exposed to the semangaka extract each with a dose of 25 mg / kg BW, 50 mg / kg BW, and 100 mg / kg BW for 20 days. The appropriate parameter is the expression of IL-6 using the Immunohistochemical method and alveolar histopathology using the Hematoxylin Eosin dye. The data analysis of IL-6 designation was done statistically quantitatively with ANOVA method taking place with BNJ (α = 0,05), whereas in alveolar histopathology was analyzed descriptively. The results showed that the prevention of watermelon (was significantly able to alter IL-6 expression and prevent alveolus histopathology. A dose of 100 mg / kgBW is the best dose capable of displaying results for the Negative Control group. The conclusion of this research is that Watermelon fruit (Citrullus lanatus) can be used as a precaution in rats which gives exposure to mosquito repellent smoke.
