Abstract

Luka bakar merupakan suatu kondisi trauma, dimana kulit mengalami kerusakan atau kehilangan jaringan secara lokal maupun sistemik. Pengobatan luka bakar secara umum dapat diobati dengan pemberian gel ekstrak plasenta karena mampu mempercepat reepitelisasi. Beberapa kejadian luka bakar menunjukkan bahwa kandungan VCO, seperti tokoferol, phytosterol, flavonoid dan asam laurat dapat digunakan sebagai terapi luka bakar. Penelitian ini bertujuan untuk mengetahui perbedaan efek terapi salep VCO dengan gel ekstrak plasenta, dalam penyembuhan luka bakar derajat II B dilihat dari peningkatan ekspresi FGF-2 dan jumlah fibroblas. Kondisi luka bakar derajat II B dibuat pada area flank dengan meletakkan solder yang telah dimodifikasi dengan plat besi berukuran 2x2 selama 10 detik. Hewan coba yang digunakan adalah tikus putih (Rattus novergicus) jantan, strain Wistar, umur 75-90 hari, dengan berat 150-200 gram. Penelitian ini bersifat eksperimental menggunakan post test only two group experimental design, dibagi dalam 2 kelompok perlakuan masing-masing 9 ekor. Kelompok 1 diberi terapi salep VCO, sedangkan kelompok 2 diberi terapi gel ekstrak plasenta. Terapi diberikan sebanyak 2 kali sehari, selama 7 hari. Perhitungan ekspresi FGF-2 dilakukan dengan membuat preparat histopatologi pewarnaan Imunohistokimia, kemudian diolah menggunakan software Immunoratio. Jumlah fibroblas diamati dengan membuat preparat histopatologi pewarnaan Hematoksilin eosin. Analisa data dilakukan secara kuantitatif menggunakan uji t. Hasil penelitian menunjukkan bahwa tidak terdapat perbedaan yang signifikan (p>0,05) antara kelompok terapi salep VCO dan kelompok terapi gel ekstrak plasenta pada kasus luka bakar derajat II B. Kesimpulan dari penelitian ini ialah terapi salep VCO pada luka bakar derajat II B tidak memberikan perbedaan efek dengan gel ekstrak plasenta

English Abstract

Burn is a trauma condition, in which skin is damaged or lost its tissue locally or systemically. The treatment of burns usually involve giving topical dosage form of placenta extract gel because it can speed up the process of reepithelization. Several incidents of burns showed that the VCO contents, like tocopherol, phytosterol, flavonoid and lauric acid could be used to treat burns. The purpose of this study was to find out the effect differences of VCO ointment therapy and placenta extract gel therapy in treating II B degree burns reviewed by the increase of FGF-2 expression and the amount of fibroblast. II B degree burn condition was made in flank area by placing a solder that has been modified with 2x2 iron plat for 10 seconds. The model animals that were being used consisted of male white Wistar rats (Rattus novergicus) age between 75-90 days and weigh around 150-200 gram. This study was experimental using post-test only two group experimental design, and the rats were divided into two groups consisted of nine rats in each group. Group 1 consisted of rats with burns that was given the VCO as the therapy, while the group 2 consisted of rats with burns that was given placenta extract gel as the therapy. Therapy was given twice a day, for seven days. The measurement of FGF-2 expression was conducted by immunohistochemistry method, and then processed using immunoratio software. The amount of fibroblast was observed through Hematoxylen-Eosin (HE) staining. The data was analyzed quantitatively using t-test to see the effect differences between two treatments. The result of this study showed that there were not any significant difference (p>0,05) between VCO ointment therapy group and placenta extract gel therapy group in treating II B degree burn cases, which indicates the VCO oinment therapy did not have a different effect with the placenta ekstrak gel therapy in treating II B degree burns.
