Abstract

Pengambilan data penelitian dimulai Desember 2012 sampai dengan Februari 2013, prosesing semen beku untuk pengujian motilitas, viabilitas dan abnormalitas dilaksanakan di Laboratorium Reproduksi Ternak Fakultas Peternakan Universitas Brawijaya dan penampungan semen segar kambing boer diperoleh dari Laboratorium Lapang Sumber Sekar. Tujuan penelitian ini adalah untuk mengetahui pengaruh glutathione terhadap kualitas semen beku kambing boer dalam medium pengencer Tris Aminomethane kuning telur serta menentukan kadar glutathione yang tepat untuk mempertahankan kualitas semen kambing boer post thawing . Metode yang digunakan dalam penelitian ini adalah metode laboratorium. Kambing boer yang diamati berumur 2-2,5 tahun dan bobot badan 80-90 kg, dengan variabel pengamatan motilitas, viabilitas dan abnormalitas terhadap perlakuan P0: 0 g glutathion e, P1: 0,2 g glutathione , P2: 0,4 g glutathione , dan P3: 0,6 g glutathione . Data yang diperoleh dianalisis dengan menggunakan Rancangan Acak Kelompok (RAK), jika didapat hasil yang berbeda nyata atau sangat nyata dari hasil statistik maka dilakukan Uji Jarak Berganda Duncan (UJBD).

English Abstract

The purpose of this study was to know the effect of glutathione in tris-yolk-glycerol extender on semen quality after rapid freezing in boer goats. Semen was collected twice a week using an artificial vagina from male goats boer aged 2 to 2,5 years old with a body weight of 85-90 kg. Semen was diluted by using tris aminomethane 10% egg yolk-7,5% glycerol 100 ml containing of 0 g, 0,2 g, 0,4 g and 0,6 glutathione/100 ml, the diluted semen was then loaded into 0,25 ml staw equilibration 5 o C for 30 minute plunged in liquid nitrogen vopar at-179 o C (4 cm above liquid nitrogen surface) and directly plunged into liquid nitrogen. After 24 hour the semen was thawed by plunging the straw into water tab 25 o C for 5 minute. Data were analyzed using a randomized block design with 4 treatments and 10 replications. Result showed that the addition of glutathione increased (P<0,05) percentage motility (73,25%, 76,45%, 77,65%, and 75,9%) and viability (82,55%, 84,65%, 86,2% and 82,7%) after dilution and is able to reduced (P<0,01) in the percentage of abnormal spermatozoa (19,16%, 14,95%, 10,9% and 21,02%) for the respective glutathione concentration and prosentase prosesing motility of spermatozoa at 0,4 g/100 ml in the diluents (77,65%, 70,25% and 20,5%), viability of spermatozoa (86,2%, 80,2% and 21,5%) and the results showed abnormalities boer goat spermatozoa were lower (10,9%, 17,86% and 20,1%) for semen processing. It can be concluded that the addition of 0,4 g glutathione resulted sperma motility and viability thawing of boer goat spermatozoa
