Abstract
Pabrik gula dalam pengolahanya selalu menghasilkan limbah logam berat Pb. Logam tersebut berasal dari bahan penjernih Pb-asetat yang digunakan untuk analisa gula dalam pengawasan pabrikasi. Bahan penjernih tersebut telah digunakan sudah cukup lama, sejak satu abad yang lalu. Diperkirakan untuk pabrik gula yang berkapasitas 4000 ton tebu per hari diperlukan tidak kurang dari 100 kg Pb per musim giling (Syahputra et al., 2011). Kurangnya manajemen sektor industri dengan ketiadaan instalasi pengolahan air limbah berpotensi menimbulkan pencemaran lingkungan terutama air sebagai sumber irigasi lahan pertanian, selain itu penggunan pupuk dari limbah tebu dapat meracuni lahan pertanian. Pencemaran Pb pada lahan pertanian menyebabkan fitotoksitas pada tanaman budidaya. Beberapa penelitian menunjukkan bahwa pencemaran unsur Pb mengakibatkan menurunnya pertumbuhan dan produksi tanaman serta diikuti dengan gejala yang tampak (visible symptoms). Unsur Pb dapat membahayakan apabila masuk ke dalam sistem metabolisme tanaman dalam jumlah yang melebihi ambang batas. Bioremediasi adalah proses detoksifikasi bahan kimia berbahaya pada lingkungan, menggunakan mikroorganisme atau tanaman fitoremediasi. Penelitian ini bertujuan untuk mengetahui potensi khamir pada limbah tebu sebagai agens bioremediasi unsur Pb. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan Jurusan Hama dan Penyakit Tanaman, Fakultas Pertanian, Universitas Brawijaya, Malang pada bulan Febuari sampai Mei 2016. Khamir diisolasi dari limbah tebu yang tercemar unsur Pb di PG. Pesantren Baru, Kota Kediri menggunakan media YEPD yang telah dicemari Pb 1 ppm. Hasil isolasi khamir yang diperoleh dari limbah tebu sebanyak tujuh isolat, dari limbah ampas tebu (bagasse) yaitu khamir Metschnikowia sp.1, Pichia sp., dan Candida sp. dari limbah blotong yaitu Metschnikowia sp. 2 dan Cryptococcus sp. dan dari limbah tetes (molase) yaitu Debaryomyces sp. dan Rhodotorula sp. Semua isolat khamir yang ditemukan diuji untuk mengetahui potensi dalam menurunkan unsur Pb. Hasil pengamatan parameter larutan uji, khamir mampu mempengaruhi derajat keasaman (pH) dan perubahahan fisik larutan dilihat dari perubahan optical dencity (OD), adanya uap air, perubahan aroma, endapan dan kekeruhan pada media uji seiring menurunya unsur Pb.
English Abstract
The sugar factory in the processing always release the heavy metal waste pb. The metal comes from the purification material Pb-acetate were used for analysis of sugar in the manufacturing supervision. The purification material has been used already for a long time, since a century ago. It is estimated that for sugar mills with a capacity of 4,000 tons of cane per day is required not less than 100 kg per season milled Pb (Syahputra et al., 2011). Lack of management of the industrial sector in the absence of a wastewater treatment plant has the potential to cause environmental pollution, especially water as a source of irrigation of agricultural land, the use of fertilizers from sugar cane waste can be toxic on agricultural land Pb pollution on agricultural land causing phytotoxicity on cultivated plants. Some studies show that the contamination of element Pb resulting in reduced growth and crop production and followed with symptoms appear (visible symptoms). Elements Pb may be harmful if it goes into the metabolic system in an amount that exceeds the threshold. Bioremediation is the process of detoxification of hazardous chemicals on the environment by using a microorganism or plant phytoremediation.This study aims to determine the potential of yeasts in sugarcane waste as bioremediation agents Pb element. Research conducted at the Laboratory of Plant Pathology Department of Plant Pests and Diseases, Faculty of Agriculture, University of Brawijaya, Malang in February until May 2016. Yeast isolated from sugarcane waste contaminated Pb element in PG. New Pesantren, Kediri City using YEPD media that has been contaminated with 1 ppm Pb. Yeast isolation results obtained from sugarcane waste as much as seven isolates, from waste bagasse that Metschnikowia sp. 1 yeast, Pichia sp., And Candida sp. of waste filter cake is Metschnikowia sp. 2 and Cryptococcus sp. and of waste drops mollase is Debaryomyces sp. and Rhodotorula sp. All yeast isolates tested were found to determine the potential in reducing heavy metals Pb. The results observation of parameter test solution, yeasts capable of affecting the degree of acidity (pH) and changes of physical solution seen from changes of optical dencity (OD) , water vapor, odor changes, sediment and turbidity in the test medium as decrease element Pb.
