Abstract

Serangan nematoda puru akar (Meloidogyne sp.) terhadap berbagai tanaman berekonomi tinggi dapat menyebabkan kerusakan secara kualitatif maupun kuantitatif. Oleh karena itu, tindakan pencegahan perlu dilakukan sebelum serangan nematoda puru akar ini semakin meluas. Penggunaan musuh alami nematoda yang berasal dari kelompok organisme, seperti jamur dan bakteri dapat digunakan sebagai agen hayati. Beberapa penelitian dilaporkan sudah banyak yang menemukan beberapa isolat bakteri yang bersifat antagonis. Penelitian ini bertujuan untuk mengetahui kemampuan isolat bakteri Pseudomonas fluorescens (UB_Pf1) dan Bacillus subtilis (UB_Bs1) dalam menyebabkan mortalitas juvenil II nematoda puru akar (Meloidogyne sp.) dan pengaruh kerapatan koloni bakteri P. fluorescens (UB_Pf1) dan B. subtilis (UB_Bs1) dalam menyebabkan mortalitas juvenil II nematoda puru akar (Meloidogyne sp.). Penelitian ini dilaksanakan di Sub Laboratorium Nematologi Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya Malang, pada bulan April sampai Juli 2014. Penelitian ini menggunakan rancangan acak kelompok (RAK) dengan 7 perlakuan dan 4 ulangan. Data hasil pengamatan dianalisis menggunakan analisis ragam pada taraf kepercayaan 95 %. Bila didapat hasil beda nyata maka dilanjutkan dengan uji BNJ taraf 5 %. Untuk perhitungan Lethal Concentration (LC50) dan Lethal Time (LT50) pada mortalitas juvenil II nematoda puru akar menggunakan analisis probit Hsin chi. Aplikasi isolat bakteri P. fluorescens (UB_Pf1) dan B. subtilis (UB_Bs1) mampu menyebabkan mortalitas juvenil II nematoda puru akar (M. javanica). Selain itu, pengaruh 3 taraf kerapatan koloni bakteri P. fluorescens (UB_Pf1) dan B. subtilis (UB_Bs1) dapat mempengaruhi mortalitas juvenil II nematoda puru akar (M. javanica). Nilai LT50 tertinggi pada aplikasi bakteri P. fluorescens dan B. subtilis kerapatan koloni 1011 cfu/ml, yaitu 32,99 jam untuk bakteri P. fluorescens dan 56,78 jam untuk B. subtilis.

English Abstract

The root knot nematode (Meloidogyne sp.) is one of important disease on some plants. This disease can reduce the quantity and quality of tubers and then causing significant yield losses. To prevent this damage, we can utilize fungi and bacteria that act as natural enemies for the nematode. Several studies had found some isolates of antagonistic bacteria. The purpose of this research are to know the ability of Pseudomonas fluorescens (UB_Pf1) and Bacillus subtilis (UB_Bs1) isolates for being the cause of second juvenile root knot nematode (Meloidogyne sp.) mortality and the effect of bacteria colonies density of P. fluorescens (UB_Pf1) and B. subtilis (UB_Bs1) to the second juvenile root knot nematode (Meloidogyne sp.) mortality. Research was conducted in Sub Laboratory of Plant Nematology, Pest and Disease Department, Faculty of Agriculture, Brawijaya University, Malang, from April to July 2014. Randomized block design with 7 treatments and 4 replications were used to conduct this research. The data were analyzed by analysis of variance and continued by HSD post hoc on 95% confidence level. Lethal Concentration (LC50) and Lethal Time (LT50) on the second juvenile root knot nematode mortality was analyzed with Hsin chi probit analysis. The application of P. fluorescens (UB_Pf1) and B. subtilis (UB_Bs1) isolates caused mortality of second juvenile root knot nematode (M. javanica). In addition, the effect of three levels of bacteria colonies density of P. fluorescens (UB_Pf1) and B. subtilis (UB_Bs1) isolates were effective to control second juvenile root knot nematode mortality (M. javanica). The highest value of LT50 was P. fluorescens (UB_Pf1) and B. subtilis (UB_Bs1) isolates with colony density 1011 cfu/ml, 32,99 and 56,78 hours, respectively.
