Abstract
Hepar merupakan salah satu organ penting yang bertanggung jawab dalam melaksanakan proses metabolisme obat. Parasetamol merupakan obat yang sering digunakan oleh masyarakat untuk penyembuhan penyakit seperti demam, pusing dll. Parasetamol tersebut dapat memberikan efek berbahaya bagi tubuh, jika dikonsumsi dalam dosis banyak dan dalam jangka waktu lama berupa kerusakan hepar. Bawang lanang (Allium Sativum L.) mengandung dua senyawa organosulfur utama yaitu y-glutamyl-S-allyl-L-cysteines dan S-allyl-Lcysteine sulfoxides (allin). Pengobatan kerusakan hepar saat ini masih menggunakan obat kimiawi yang memiliki efek samping. Hal ini menjadikan peluang tanaman tersebut dikembangkan menjadi ekstrak untuk kerusakan hepar. Tujuan dari penelitian ini adalah untuk mengetahui efektivitas pemberian ekstrak Bawang lanang terhadap pencegahan kerusakan hepar dan penurunan kadar SGPT dan SGOT pada tikus wistar jantan. Penelitian ini menggunakan metode penelitian RAK dengan 7 kelompok perlakuan. Data yang diperoleh dianalisis dengan Analisis Varian (ANOVA) dan apabila terjadi perbedaan antar perlakuan, maka dilanjutkan dengan uji beda nyata yaitu BNT (Beda Nyata Terkecil) dengan selang kepercayaan a=5% Pada pengujian bahan ekstrak bawang lanang didapatkan nilai aktivitas antioksidan (IC50) 65,074 g/mL total fenol 544,3 (mg GAE/g), dan total flavonoid 218,3 (mg quarcetin/g). Hasil pengujian in vivo menunjukkan pengaruh beda nyata (a=0,05) terhadap penurunan SGPT pada tikus sehat, SGPT dan SGOT pada tikus yang diinduksi paracetamol. Pengaruh tidak beda nyata (a>0,05) terhadap penurunan SGOT pada tikus sehat. Pemberian esktrak dosis 15 mg/kg BB dipilih sebagai perlakuan terbaik.
English Abstract
Hepar is one of important organs responsible in drugs metabolism. Paracetamol commonly used by people to cure diseases such as a fever, dizzy, etc. . Paracetamol can provide harmful effects to the body such as hepatic damage, if consumed in large doses and in the long term. Lanang garlic (Allium Sativum L.) containing two major organosulfur compounds which are y-glutamyl- S-allyl-L-cysteines and S-allyl-L-cysteine sulfoxides (allin). Damage treatment of hepar is still use chemical medicine has side effects Treatment of liver damage is still using chemical medicine that have side effects. It makes opportunity the plant developed into extract for hepatic damage. The purpose of this research is to determine the effectiveness of lanang garlic extract on the to prevention of hepatic damage and decreased levels of SGPT and SGOT in male wistar rats. This research used CRD method with 7 treatment groups. The data obtained were analyzed by Variant Analysis (ANOVA) and if there was a difference between the treatments, then continued with the real difference test which is using SRD (The Smallest Real Difference) with the confidence interval a=5%. The results from the lanang garlic extract, was found that the activity of antioxidant (IC50) 65,074 (pg/Ml),total phenol 544,3 (mg GAE/g), and total flavonoids 218,3 (mg quercetin/g). In vivo test results showed significant difference (a=0,05) to SGPT reduction in healthy rats, and degradation of SGPT and SGOT value on induced paracetamols rats. The effect was not significantly different (a>0,.05) on the SGOT decrease in healthy rats. Supplementation of the extract of dose 15 mg / kg BW was chosen as the best treatment.
