Abstract
Tembakau merupakan komoditas perkebunan yang bernilai ekonomi tinggi, sebagai sumber penerimaan negara melalui devisa, cukai dan pajak. Tembakau di beberapa sentra produksi merupakan sumber pendapatan petani, penyedia lapangan kerja dan penggerak ekonomi di daerah. Tembakau digunakan sebagai bahan baku rokok dan juga sebagai tembakau kunyah atau susur. Salah satu penyakit penting yang menyerang tanaman tembakau di Indonesia adalah penyakit lanas yang disebabkan oleh jamur Phytophthora nicotianae. Pengendalian P . nicotianae sulit dilakukan karena P . nicotianae dapat bertahan hidup di dalam tanah sebagai saprofit fakultatif jika tidak ada inang. Pengendalian menggunakan fungisida mempunyai dampak negatif terhadap lingkungan, mikroorganisme nontarget, terjadinya resistensi patogen, dan menurunnya keanekaragaman mikroorganisme tanah serta biaya untuk pembelian fungisida cukup tinggi. Pengendalian dengan menggunakan agensia hayati lebih aman dan ramah lingkungan meskipun hasilnya tidak seefektif fungisida. Tujuan penelitian ini adalah untuk mengetahui masa simpan bakteri antagonis Bacillus sp. dalam media cair yang masih efektif mengendalikan penyakit lanas P. nicotianae pada tanaman tembakau. Penelitian ini dilakukan di Laboratorium Fitopatologi dan rumah kaca Balai Penelitian Tembakau dan Tanaman Serat(BALITTAS) Karangploso Malang. Penelitian dilaksanakan pada bulan Oktober 2010 sampai Januari 2011. Penelitian terdiri dari dua tahap penelitian yaitu secara in vitro dan secara in vivo . Penelitian secara in vitro terdiri dari uji Dual Culture 2 titik, uji Dual Culture 5 titik dan uji Pour Plate . Penelitian secara in vivo yaitu uji intensitas serangan penyakit lanas P . nicotianae pada tanaman tembakau. Tahap in vitro terdiri dari 10 perlakuan, 2 kali ulangan. Sedangkan tahap in vivo terdiri dari 4 perlakuan, 3 ulangan setiap ulangan terdiri dari 9 sampel tanaman tembakau. Hasil data tahap in vitro maupun in vivo dianalisis menggunakan uji t 5 %. English Abstract
Tobacco is a high economic value commodity, hence it has a function as a source of state revenues through foreign exchange, customs and taxes. Tobacco in some production center is a source of income of farmers, employers and economic drivers in the area. Tobacco is used as raw materials as well as cigarettes and chewing tobacco or fringe. One of the important diseases that attack tobacco Sweet is blackshank caused by the fungus Phytophthora nicotianae . Control P. nicotianae is difficult because P. nicotianae can survive in soil as a facultative saprophyte in the absence of the host. The us of fungicides has negative impact on the environment, non target microorganisms, causes emergence of resistant pathogens, and decreases diversity of soil microorganisms as well as the costs for the purchase of fungicides is quite high. Using biological control agents are safer and environmentally friendly, but the results are not as effective as fungicides. The purpose of this study was to determine the effectiveness of antagonistic bacterium Bacillus sp. in a liquid medium against P. nicotianae on tobacco plants. The bacterium was stored in various periods, ie. 3, 6, and 12 months. The research was conducted in laboratory and greenhouse Fitopatologi Research Institute for Tobacco and Fiber Crops (BALITTAS) Karangploso Malang. The experiment was conducted in October 2010 to January 2011. The study consisted of two phases of activity, i.e. in vitro and in vivo . In vitro test consisted of Dual Culture using 2 spots, Dual Culture test using 5 spots, and Pour Plate test. In vivo test determind the black shank severity on tobacco plants. In vitro consisted of 10 treatments, two replicates. While the in vivo test consisted of 4 treatments, 3 replicates with each traetment consisted of nine samples of tobacco plant. The data were analyzed using the t test 5%. The results of in vitro test in Dual Culture 2 points, Dual Culture 5 points and Pour Plate test showed that Bacillus sp.
