Abstract
Diabetes Melitus (DM) tipe 1 adalah penyakit metabolisme yang mempunyai karakteristik terjadi hiperglikemia (kadar gula darah meningkat) akibat kerusakan sel beta pankreas yang menyebabkan penurunan produksi insulin. Albedo semangka (kulit buah yang berwarna putih mengandung zat sitrulin sebesar 60% Zat sitrulin akan bereaksi dengan enzim tubuh ketika dikonsumsi, lalu diubah menjadi arginin yang merupakan asam amino nonesensial yang memiliki peran sebagai antioksidan untuk melawan radikal bebas pada tubuh. Tujuan penelitian ini untuk menguji ekstrak albedo semangka merah sebagai terapi Diabetes Melitus tipe 1 yang diinduksi Streptozotocin. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Penelitian menggunakan tikus (Rattus norvegicus) jantan berusia 8-12 minggu, berat badan 100-150 gram terdiri dari lima kelompok, yaitu: kontrol negatif, kontrol positif, P1 ekstrak albedo semangka merah 500 mg/kg BB, P2 1000 mg/kg BB dan P3 1500 mg/kg BB. Pemeriksaan parameter yaitu histopatologi ginjal dengan pewarnaan HE dan Ekspresi IL-1β metode Imunohistokimia dan dianalisis secara statistik dengan one-way ANOVA, (α=0,05) dan uji Tukey. Hasil penelitian menunjukkan pemberian ekstrak albedo semangka merah menurunkan ekspresi IL-1β dan kerusakan ginjal dengan dosis terbaik yaitu 1500 mg/Kg BB. Kesimpulan penelitian ini, ekstrak albedo semangka merah dapat digunakan terapi bagi penderita Diabetes Melitus tipe I
English Abstract
Diabetes mellitus type 1 (DM) is a metabolic disease that had characterized happens hyperglycemia (increased blood sugar levels) due to pancreatic beta cell damage that will cause the onset of insulin deficiency. Watermelon albedo (whiterind contain a sitrulin of 60% sitrulin Substances will react with the body's enzymes when consumed, and then converted into arginine which is a nonessential amino acid that has a role as an antioxidant to fight free radicals in the body. The purpose of this research was to test the Red watermelon albedo extracts as a therapy for Diabetes mellitus type 1 Streptozotocin induced. This experiment is going to designed use completely randomized design. Research using rats (Rattus norvegicus) males aged 8-12 week, weight 100-150 grams divided five groups, namely: the negative control, positive control, P1 red watermelon albedo extract 500 mg/kg, P2 1000 mg/kg and P3 1500 mg/kg. Examination of the parameters, namely kidney histopathology with HE staining and expression of IL- 1 β Immunohistochemical method and analyzed statistically with ANOVA, oneway (α = 0.05) and Tukey test. The results showed the awarding of the Red watermelon albedo extracts decreases the expression of IL-1 β and kidney damage with a dose of 1500 mg/Kg BB. Conclusions this study, red watermelon albedo extracts can be used in therapy for patients with Diabetes mellitus type I.
