Abstract
Oviduct Specific Glycoprotein (OVGP) diketahui dapat meningkatkan fisiologis dan fluiditas membran serta membantu peningkatan ion kalsium ke dalam membran yang merangsang ikatan membran dengan cAMP intraseluler sehingga meningkatkan integritas membran utuh spermatozoa. Melihat potensi dari OVGP tersebut maka dilakukan penelitian tentang pengaruh suplementasi OVGP pada pengencer semen kambing yang diharapkan dapat digunakan dalam pembuatan semen beku dan mengurangi kejadian polispermi sehingga membantu meningkatkan populasi ternak kambing di Indonesia. Penelitian ini dilakukan untuk mengetahui ekspresi OVGP pada testis kambing dan pengaruh suplementasi OVGP pada pengencer semen kambing terhadap integritas membran spermatozoa kambing. Pengamatan ekspresi OVGP pada testis dilakukan dengan metode imunohistokimia dan pengamatan integritas membran dilakukan dengan membagi media pengencer spermatozoa menjadi tiga kelompok perlakuan yaitu kelompok P0 sebagai kontrol (tanpa suplementasi OVGP), kelompok P2 (OVGP dosis 5 μg/mL), dan kelompok P2 (OVGP dosis 10 μg/mL) dengan metode pemeriksaan HOS Test. Hasil penelitian menunjukkan ekspresi OVGP pada testis kambing terlihat pada bagian sel-sel basis dari tubulus seminiferus yaitu sel spermatogonium dan sel spermatosit serta pada sel interstitial yaitu sel leydig. Suplementasi OVGP berpengaruh terhadap integritas membran secara signifikan (P<0,05) pada dosis 5 μg/mL dan 10 μg/mL dibandingkan dengan tanpa suplementasi OVGP. Berdasarkan hasil tersebut dapat disimpulkan bahwa terdapat ekspresi OVGP pada organ testis kambing serta terdapat peningkatan integritas membran spermatozoa yang disuplementasi oleh OVGP pada pengencer semen kambing.
English Abstract
Oviduct Specific Glycoprotein (OVGP) is known to improve physiological and membrane fluidity and helps increase calcium ions into membranes that stimulate membrane bonding with intracellular cAMP thus enhancing the integrity of intact membranes of spermatozoa. Considering the potential of OVGP, research on the effect of OVGP supplementation on goat cement dilution is expected to be used in the manufacture of frozen semen and reduce the incidence of polispermi thus helping to increase the population of goats in Indonesia. This study was conducted to determine the expression of OVGP in goat testis and the effect of OVGP supplementation on goat cement diluent on goat spermatozoa integrity membrane. Observation of OVGP expression in the testis was done by immunohistochemical method and membrane integrity observation was done by dividing the spermatozoa dilution medium into three treatment groups ie group P0 as control (without OVGP supplementation) (50,31 ± 3,37), group P2 (OVGP dose 5 μg / mL) (53.12 ± 1.57), and P2 group (OVGP dose 10 μg / mL) (54.81 ± 1.64) with HOS Test method. The results showed that OVGP expression in goat testis was seen in the base cells of seminiferous tubules of spermatogonium and spermatocyte cells and in interstitial cells of leydig cells. OVGP supplementation significantly affected membrane integrity (P <0.05) at doses of 5 μg / mL and 10 μg / mL compared with no OVGP supplementation. Based on these results it can be concluded that there is an expression of OVGP in goat testis organ and there is increased integrity of membrane spermatozoa supplemented by OVGP on goat cement diluent.
