Abstract
Luka insisi yaitu suatu keadaan yang ditandai dengan rusaknya jaringan karena irisan benda bertepi tajam, seperti pisau, silet, dan sejenisnya. Apel Manalagi (Malus sylvestris Mill) memiliki kandungan zat aktif yaitu flavonoid sebagai anti inflamasi dan vitamin c dalam membantu proses regenerasi jaringan, sehingga mempercepat waktu penyembuhan dengan pembentukan kolagen pada jaringan ikat, pembentukan membran basalis dan matriks antar sel. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian salep ekstrak ampas apel Manalagi (Malus sylvestris Mill) terhadap ekspresi interleukin-6 (IL-6) dan gambaran sel radang luka insisi selama proses penyembuhan luka. Hewan coba yang digunakan 20 ekor tikus putih strain wistar umur 8-12 minggu, dengan berat badan 150-250 gram, dibagi 5 kelompok yaitu kontrol negatif, kontrol positif, perlakuan terapi salep ekstrak ampas apel Manalagi masing masing konsentrasi 25%, 35%, dan 45%. Data kuantitatif dianalisa statistik ANOVA dilanjutkan uji Tukey α = 0,05. Hasil penelitian menunjukkan bahwa salep ekstrak ampas apel manalagi secara signifikan (p<0,05) menurunkan ekspresi IL-6 dan menurunakan jumlah sel radang pasca luka insisi. Kesimpulan dari penelitian ini yaitu salep ekstrak ampas apel manalagi konsentrasi 45% mampu dalam mendukung proses penyembuhan luka insisi.
English Abstract
Incision wound is a condition where the continuity of tissues is damaged by trauma of sharp object, such as a knife, a razor blade, and the others. Manalagi Apple (Malus sylvestris Mill) contain an active substance which is flavonoid as an anti-bacterial and vitamin C for regeneration of tissue, thus the recovery with formation of the collagen in connective tissue, the formation of basal membranes and matrix between cells. The aim of this research is to know the effect of apple pulp Manalagi (Malus sylvestris Mill) ointment on expression of interleukin-6 (IL-6) and incision inflammatory cell image during wound healing process. The research subjects were twenty male rats wistar strain aged 8-12 weeks, weight 150-250 gram divided to five groups : negative control, positive control, experimental group (rats with incision wound and given extract apple pulp ointment each concentrations are 25%, 35%, and 45%). The results of IL-6 expression were observed using immunohistochemistry and the amount of inflammatory cells using HE staining. Quantitative data analysis of interleukin 6 (IL-6) expression and inflammatory cell expression using one-way ANOVA test and if there is a significant difference followed by Tukey test α = 0.05. This result of this study discovered that extract apple pulp ointment therapy significantly (p<0,05) decrease IL-6 expression and decrease inflammatory cells. The conclusion of this study is extract apple pulp manalagi each concentrations 45% can be used as an good treatment for incision wound healing.
