Abstract
Glomerulonefritis Akut (GNA) merupakan penyakit kompleks imun ditandai dengan inflamasi dan proliferasi sel glomerulus pada ginjal, Streptokinase diproduksi oleh streptococcus beta hemoliticus yang dapat menyebabkan glomerulonefritis akut. Tujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak daun sukun (Artocarpus Altilis) sebagai terapi pada glomerulonefritis akut berdasarkan penurunan kadar MDA dan perbaikan gambaran histopatologi ginjal. Penelitian ini menggunakan model Rancangan Acak Lengkap (RAL). Mencit dibagi ke dalam 5 kelompok yaitu, kelompok kontrol negatif, kontrol positif, dan 3 kelompok perlakuan yang diinduksi streptokinase secara IV dosis 2500 IU dan diterapi dengan ekstrak daun sukun (dosis 16,8 mg/ekor, 25,2 mg/ekor, dan 33,6 mg/ekor per-oral selama 14 hari. Analisis kuantitatif kadar MDA menggunakan Oneway ANOVA dengan (P<0.05).Analisa kualitatif digunakan untuk menganalisis gambaran histopatologi ginjal. Hasil penelitian menunjukkan pemberian terapi ekstrak daun sukun dengan dosis 16,8 mg/ekor, 25,2 mg/ekor dan 33,6 mg/ekor mampu menurunkan kadar MDA dan memperbaiki gambaran histopatologi ginjal yaitu perbaikan pada gambaran ginjal dengan penurunan infiltrasi sel radang, perbaikan ukuran kapsula bowman dan perbaikan tubulus yang ditunjukkan dengan penurunan nekrosis pada epitel tubulus. Kesimpulan penelitian ini yaitu ekstrak daun sukun dapat sebagai terapi pada mencit model glomerulonefritis akut hasil induksi streptokinase berdasarkan penurunan kadar MDA dan perbaikan gambaran histopatologi ginjal dengan dosis terbaik yaitu 16,8 mg/ekor.
English Abstract
Glomerulonephrithis Acute is an immune complex disease in kidney that mark by inflammation and proliferation of glomerulus cell. Streptokinase made from streptococcus beta hemoliticus causing glomerulonefrithis acute. The purpose of this study is to know the effect of the extract leaves of sukun as therapy effect of glomerulonephrithis acute based on decrased MDA level and repair of histopathology ren. This experiment use completely randomized design (CRD). Mice were divided into 5 groups: negative control group, positive control group, and three groups of IV induction of streptokinase dose 2500 IU and treated with (Artocarpus Altilis) (Dose 16,8 mg/mice, 25,2 mg/mice, and 33,6 mg/mice per-oral for 14 days). Quantitative analysis of MDA level using One Way ANOVA with (P <0.05). Qualitative analysis was used to analyze the histopathology of ren. The result showed that extract leaves of Artocarpus altilis with doses 16,8 mg/mice,25,2 mg/mice and 33,6 mg/mice has significanly therapy effect to glomerulonephrithis acute based on decrease levels of MDA in ren and repair of histopathology ren that is repair of ren by decrease of radang infiltration, repair size of Capsula Bowman, and repair tubulus by decrease necrosis of epitel tubulus. The conclusion of this study was Artocarpus altilis has therapy effect to glomerulonefrithis acute induced by streptokinase based on decrease levels of MDA in ren and repair of histopahatology ren with the best dose is 16,8 mg/mice.
